摘  要:目的 采用温度固化型聚氧化乙烯水凝胶作为骨髓基质干细胞 (MSCs)的注射型载体 ,观察水凝胶-细胞复合物注入裸鼠体内后骨组织的形成情况。方法 体外培养并扩增兔MSCs,将细胞按 5× 10 7/ml接种于聚氧化乙烯溶液中 ,形成凝胶后注射于裸鼠背部皮下组织中 ,于注射后 1,2个月取材 ,通过大体标本观察、组织学检查 ,观察骨组织的形成情况。结果 注射后 1个月 ,在皮下形成稍硬的结节 ,2个月时形成的结节质地坚硬 ,外观为骨组织 ;1个月标本中可见大量的软骨组织和类骨组织 ,2个月标本中有成熟的骨组织形成。结论 温度固化型聚氧化乙烯水凝胶作为成骨细胞载体 ,可以通过注射的方法再造骨组织 ,具有广阔的应用前景。Objective To detect temperature-dependent polymerising polyethylene oxide hydrogel used as a vehicle to deliver bone marrow stem cells(MSCs) and to observe new bone regeneration activity of this cell-hydrogel complex.Methods MSCs were harvested by cell culture,and cells were seeded into polyethylene oxide solution.The density of the suspension was adjusted to 5×10 7/ml.The hydrogel was obtained by adjusting the temperature to over 6℃.Aliqots of 0.5ml of the cell hydrogel complexes were injected subcutaneously into the backs of the experimental mice,and 0.5ml of hydrogel alone was injected as the controls.Regeneration of new bone was studied grossly and histologically.Results The 1st month after injection hard nodus formed subcutaneously,and the 2nd month after injection the nodus became harder and showed the appearance of new bone.By histological observation,woven bone was seen in the one month specimens,and in some area cartilage could also be observed.In the two months specimens mature new bone could be observed.Conclusion New bone could be regenerated through the injection of temperature-dependent polymerising polyethylene oxide hydrogel seeded with MSCs,this method might have great potential of clinical use in the near future.
