摘 要:目的 观察以AnnexinV/PI法检测新风胶囊对佐剂性关节炎 (adjuvantarthritis,AA)大鼠滑膜、胸腺及胃黏膜细胞凋亡的影响 ,探讨新风胶囊的作用机制。方法 采用弗氏完全佐剂造成佐剂性关节炎 (AA)大鼠模型 ,设立正常对照组、模型对照组、甲氨喋呤 (MTX)对照组、雷公藤多苷AIM To observe the effect of xinfeng capsule (XFC) on the apoptosis of the synoviocyte, thymus lymphocyte and gastric mucosa cell of adjuvant arthritis (AA) rat by Annexin V/PI. METHODS Model rats were established by the Freund's complete adjuvant. Fifty rats were randomly divided into 5 groups: normal control, model control, methotrexate (MTX) control, triperygium wilfordii polycoride tablet (TPT) control, xinfeng capsule treated (XFC). The arthritisindex ( AI ) and weight of rats was recorded.The apoptotic percentage of the synoviocyte? thymus lymphocyte and gastric mucosa cell of AA was measured by flow cytometry (FCM) using Annexin V/PI. RESULTS Compared with before treatment, XFC, MTX and TPT can decrease the AI (P<0 01~0 05) , the weight of rats in XFC increased markedly after treatment (P<0 05 ) and the weight of rats in TPT and MTX had no marked change (P>0 05) . Compared with the normal control, apoptotic percentage of the synoviocyte and thymus lymphocyte of model control was decreased and apoptotic percentage of gastric mucosa cell of model control was increased (P<0 05) ; Compared with the MTX, TPT and model control, apoptotic percentage of the synoviocyte and thymus lymphocyte of XFC was significant increased and apoptotic percentage of gastric mucosa cell of XFC was decreased ( P<0.05~0 01). CONCLUSION XFC can decrease the AI like MTX, TPT. But it is superior to MTX, TPT in increasing the weight of rats and the apoptosis of the synoviocyte and thymus lymphocyte, decreasing the apoptosis of the gastric mucosa cell of AA. It maybe the mechanism for XFC that promot the apoptosis of synoviocyte and thymus lymphocyte, restrain the synovial membrane from hyperplasia and autoimmune reaction, restrain the apoptosis of gastric mucosa cell and protect gastric mucosa.
