摘  要:目的 探讨彩色多普勒超声照射对中孕期胎鼠肾小管上皮细胞bcl 2蛋白表达的影响。方法  3 2只孕 14天SD大鼠按彩超辐照时间不同随机等分为四组 ,Ⅰ组 (对照组 )、Ⅱ组 (照射 10min组 )、Ⅲ组 (照射 2 0min组 )、Ⅳ组 (照射 3 0min组 ) ,仪器采用Acuson公司Sequoia 5 12型彩色电脑声像仪 ,照射条件 :4V1探头 ,二维频率 3 .0MHz ,彩色频率 3 .0MHz ,Tis =1.8,MI =1.6。照射后 2 4h剖宫取胎鼠肾脏标本 ,HE染色光镜观察组织结构 ;免疫组化ABC法检测肾脏bcl 2蛋白的表达 ,采用LeicaQ 5 0 0MC型图像分析仪进行图像分析。结果 Ⅰ~Ⅳ组光镜下观察肾脏组织结构均未见异常 ;免疫组化法检测 ,各组胎鼠肾小管上皮内可见均匀散在的棕黄色淡染的bcl 2蛋白阳性表达 ,图像分析统计结果显示 :Ⅱ组及Ⅲ组胎鼠肾小管上皮bcl 2蛋白阳性表达强度与Ⅰ组无显著差异 (P >0 .0 5 ) ;Ⅳ组表达强度与Ⅰ组有显著差异 (P <0 .0 5 )。结论 彩色多普勒超声照射中孕期大鼠超过 3 0min可引起胎鼠肾小管上皮细胞bcl 2蛋白表达减少。Objective To study the effect of diagnostic CDFI on the expression of bcl 2 protein in fetal rat's renal tubule epithelial cell. Methods Thirty two female Spraque Danles rats that have been pregnant for 14 days were divided into four groups averagely and randomly, control group (groupⅠ) was sham irradiation, groupⅡ Ⅳ were irradiated by diagnostic CDFI (Sequoia-512, 4V1, 3.0 MHz, Tis=1.8, MI =1.6) for 10 min, 20 min and 30 min respectively, fetal rats' kidneys of four groups were removed at 24 h after irradiation, HE stain was done for microscope observing, expression of bcl 2 protein was detected with immunohistochemical method. Results There was no abnormal changes among group Ⅰ Ⅳ under microscope, and there was no significant differences in expression of bcl 2 protein among group Ⅰ,group Ⅱ and group Ⅲ ( P >0.05). But there was significant difference in expression of bcl 2 protein between groupⅠand group Ⅳ ( P < 0.05 ). Conclusion Continuous irradiation beyond 30 minutes by diagnostic CDFI may result in decreasing expression of bcl 2 protein in fetal rat's renal tubule epithelial cell.
