中药复方搏癌丸对人肝癌细胞周期及bax、bcl-2、p53基因表达调控的影响

摘　　要：目的 :研究中药复方搏癌丸对人肝癌细胞HepG2周期及相关调控基因表达的影响。 方法 :采用药物血清和细胞药理学方法 :①用流式细胞仪观察搏癌丸对人肝癌HepG2细胞凋亡及周期的影响。②用免疫组织化学染色(SABC)法检测搏癌丸 3个有效药物血清浓度对bax、bcl 2、p5 3基因表达的调控。 结果 :① 10 %搏癌丸药物血清作用HepG2细胞 48小时后 ,FCA检测的细胞凋亡率为 2 5 3 4% ,均显著高于相应无药血清对照组的 3 5 4% ,P <0 0 5。②能上调HepG2细胞凋亡调控基因bax、p5 3的表达 ,与无药血清对照组比较 ,差异有显著性意义 ,P <0 0 5 ,且呈剂量依赖关系。结论 :搏癌丸诱导HepG2细胞凋亡的作用机制之一可能是上调促凋亡基因bax和 p5 3的表达。Objective:To study the effects of Boaiwan mixture on hepatocellular cancer line HepG2 in cell cycle and related gene expression. Methods:The apoptosis and cell cycle of hepatocellular cancer cell were studied by flow cytometry. The expression of bax gene?bcl-2 gene and p53 gene were examinated by immunohistochemical staining method after treated with three efficient concentration of medicated serum containing Boaiwan. Results:10% medicated serum induced apoptotic rate was 25.34% by 48h while the control group was 3.54%. There was a significant difference,P<0.05. Boaiwan mixture can up-regulate the expression of bax gene and p53 gene. It was significantly different from control group (P<0.05) and the effects revealed a medicated serum concentration dependent manner. Conclusion:One of the mechanisms of human hepatocellular cancer cells by Boaiwan mixture is to up-regulate the expression of bax gene and p53 gene.
