柴葛口服液中葛根素在犬体内的药动学及绝对生物利用度

摘　　要：目的 :研究柴葛口服液中主要成分葛根素在犬体内的药物动力学及绝对生物利用度。方法 :家犬 6只 ,随机分为 2组 ,采用单剂量交叉给药方案 ,分别给予单剂量口服柴葛口服液和静脉注射葛根素注射液 ,用HPLC法测定血药浓度 ,并用3p97程序拟合药动学参数。结果 :柴葛口服液在家犬体内的药 时数据符合二室模型 ,主要药动学参数为t1/2 (β) =14 .84min ,CL =16 .4 8ml·h-1·kg-1,Cmax=2 5 .74 μg·ml-1,Tmax=36 .2 4min ,AUC(0→∞ ) =32 812 9.0 5 μg·ml-1·min ,F =4 0 .4 3%。结论 :柴葛口服液的生物利用度较好。Objective: To study the pharmacokinetics and absolute bioavilability of puerarin in Chaige oral liquid in sera of dogs.Method: A single dose of Chaige oral liquid or puerarin were orally or iv given in 6 dogs by an open randomized crossover test. HPLC was selected to determine the concentration of puerarin in serum.The pharmacokinetic parameters of Chaige oral liquid was caculated and analysed by 3P97 program.Result: The data of durg time of puerarin was accord with two compartment model.The t 1/2(β) ,CL, C max , t max , AUC (0→∞) and absolute bioavailability of Chaige oral liquid were 14.84 min, 16.48 ml·h -1 ·kg -1 , 25.74 μg·ml -1 , 36.24 min, 328129.05 μg·ml -1 ·min,and 40.43% , respectively.Conclusion: The bioavailability of puerarin of Chaige oral liquid was mediate.
