环孢素A对人Tenon’s囊成纤维细胞增殖及TGF-β_1分泌的影响

摘　　要：目的　观察环孢素A(CsA)对体外培养的人Tenon’s囊成纤维细胞 (HTFs)增殖及其转化生长因子 β1 (TGF β1 )分泌的影响。方法　以体外培养的人HTFs为实验对象 ,分别加入 0 0 0 1～ 10 μg/mLCsA作用 48h ,细胞计数观察CsA对细胞增殖的影响。收集 0 0 0 1～ 5 μg/mLCsA处理后细胞培养上清液 ,采用ELISA方法检测细胞TGF β1 的分泌量。光镜及透射电镜观察细胞的结构变化。结果　CsA在 0 1～ 10 μg/mL范围内以质量浓度依赖方式抑制人HTFs的增殖 ,IC50 为0 5 5 μg/mL ,并在 0 0 1～ 5 μg/mL范围内刺激细胞TGF β1 的分泌。CsA作用后细胞形态学改变的主要特征为胞浆内大量空泡形成。结论　一定质量浓度的CsA可能通过直接的细胞损伤作用在促进TGF β1 分泌的同时仍显著抑制人HTFs的增殖。Objective To explore the effects of cyclosporin A(CsA) on proliferation and TGF-β 1 production of cultured human Tenon’s capsule fibroblasts (HTFs). Methods Effect of CsA(0.001 μg/mL10 μg/mL)on proliferation of cultured HTFs was studied by cell counting after 48-hour exposure.Overlying growth medium was collected after exposure to 0.001 μg/mL5 μg/mL of CsA for 48 hours,and the production of transforming growth factor-β 1(TGF-β 1) was quantified using enzyme-linked immunosorbant assay(ELISA).The morphological changes of HTFs was observed with light microscope and transmission electron microscope. Results 0.1 μg/mL10 μg/mL of CsA inhibited HTFs proliferation in experimental groups in comparison with the control group(P<0.05)in a dose-dependent manner,and the drug concentration meeting for 50% inhibition of growth was 0.55 μg/mL. 0.01 μg/mL5 μg/mL of CsA stimulated the production of TGF-β 1 in the experimental groups compared with the control group(P<0.05).The morphological changes of cells after CsA exposure consisted of the presence of numerous cytoplasmic vacuoles under light microscope and the toxic changes of mitochondrion,rough endoplasmic reticulum and Golgi complex under transmission electron microscope. Conclusion While stimulating TGF-β_ 1 production,CsA can significantly inhibit cultured HTFs proliferation through direct cell injury.
