摘 要:本实验以野生滇南杜鹃种子为试验材料,通过组培的方法探究滇南杜鹃种子无菌播种与快繁技术体系。结果表明:最佳消毒方法为:70%酒精浸泡30 s + 0.1%升汞浸泡3 min (2次),然后接种于WPM + 4 mg/L ZT + 0.01 mg/L NAA培养基中,滇南杜鹃种子萌发率为76.67%、污染率16.67%。诱导不定芽的最佳培养是WPM + 2 mg/L ZT + 0.01 mg/L NAA,30 d后每株平均分化芽数为2.92,苗叶色深绿,枝条粗壮,丛生芽增多。较好的生根培养基是WPM + 1 mg/L NAA + 1 mg/L IBA,30 d统计生根率为57%,平均生根数是3.21,根长势最好,形态正常、生长整齐。研究结果为滇南杜鹃的扩大繁殖和资源保护提供了参考。In this experiment, the technique system of aseptic seeding and rapid propagation of wild Rhododendron hancockii Hemsl. seeds was studied by tissue culture. The results showed that the best disinfection method was: 70% alcohol soaked in 30 s + 0.1% mercury for 3 min (twice);then inoculated in the medium of WPM + 4 mg/L ZT + 0.01 mg/L NAA, the germination rate of R. hancockii Hemsl. seeds was 76.67% and the contamination rate was 16.67%. Induction of adventitious buds of the best culture was WPM + 2 mg/L ZT + 0.01 mg/L NAA. After 30 days per plant, average number of bud formation was 2.92;seedling leaf was dark green;cespitose bud increased. The best rooting medium was WPM + 1 mg/L NAA + 1 mg/L IBA, the rooting rate of 30 d was 57%, the average rooting number was 3.21, and the root grew best, and the shape was normal and the growth was regular. The results provide reference for expanding propagation and resource conservation of Rhododendron hancockii Hemsl.
