摘 要:目的:探讨两种纳米羟基磷灰石/硫酸钙复合材料的体外降解特点、细胞毒性,进而验证该种复合材料的细胞相容性;方法:按照n-HA在复合材料中所占质量分数不同分为复合材料A (10% n-HA)和复合材料B (20% n-HA),以磷酸盐缓冲液PBS为溶剂检测材料体外降解性能;通过MTT实验评定复合材料的细胞毒性;将人成骨样细胞MG-63种植在材料表面,然后扫描电镜观察。结果:体外降解实验结果显示A组材料在PBS缓冲液中有较明显的快速失重期和慢速失重期,第40天完全降解;B组材料没有明显的快速失重期,第25天降解至50%,第55天降解完全。MTT实验结果显示两组复合材料不同浓度的浸提液对细胞的生长、增殖没有明显影响,MTT测试得出的结果为两复合材料的细胞毒性为1级,即无细胞毒性,并两种材料差别无统计学意义(P 】0.05);两组材料与人成骨样细胞MG-63共培养的扫描电镜显示,细胞的形态、生命活动未受明显影响。结论:1) B组材料体外降解速度较A组材料慢,后者降解初期有一个短暂的快速失重期,B组材料降解相对平稳;2) A组材料与B组材料对人成骨样细胞MG-63的增殖无明显影响,细胞毒性均为1级,即无细胞毒性;细胞在两组材料表面覆盖面积均在80%以上,两复合材料均具有良好的细胞相容性。Objective: Two composites were tested in vitro in order to study their cell compatibility. Methods: Composite A (10% n-HA) and composite B (20% n-HA) were shaped into 5 mm ×3.3 mm cylinders. Properties of degradation in vitro were studied through soaking the composites into PBS buffer so-lution. In order to study two composites’ cell compatibility, MTT Test were carried out by culturing MG-63 cells in the extraction of the composites. Then MG-63 cells were seeded on the top surfaces of the composites, cellular morphologic studies were implemented by SEM (scanning electron micro-scope). Results: There was a rapid period and slow period of weight loss for composite A during the vitro degradation experiment. Composite A depredated completely at the 40th day. Composite B had no rapid weight loss period, the point of 50% weight loss was at the 25th day and complete degrada-tion was achieved at the 55th day. The results of two composites’ MTT Test showed no cytotoxicity of them. The cytotoxicity grades for both composites were Grade 1. No statistic differences between two composites (P >0.05). The SEM results showed that MG-63 cells’ life activity and cellular mor-phology stayed in normal. Conclusions: 1) Composite A degradated faster than composite B in vitro, Composite A had a period of rapid weight loss, while composite B degradated smoothly during the experiments. 2) There was no obvious influence for both composites on the proliferation of human osteosarcoma cells MG-63, MG-63 cells perform normally on the surface of both two composites, composite A and composite B showed good cell compatibility in the study.
