硫酸铜对斑马鱼发育毒性和运动能力的影响

摘  要:目的:完成研究内容,测定不同浓度硫酸铜对斑马鱼发育毒性和运动能力的影响,阐述斑马鱼对环境水体铜离子压力的敏感程度。方法:第0天收集鱼卵,第3天配制药,第3天开始给药至第7天。空白组,低剂量组,中剂量组,高剂量组,每组三个平行实验组。从给药开始拍照记录。统计幼鱼存活率。实验中需要应用恒温、恒湿和程序光照控制培养体系培育斑马鱼,鱼卵收集和孵化孵化箱、体式显微镜和显微镜及运动跟踪摄像、分析系统,显微镜观察斑马鱼形态变化。结果:本实验中硫酸铜会对斑马鱼幼鱼造成全身性损伤且96 h实验组中均有斑马鱼幼鱼死亡,0.5 μg/mL的组中斑马鱼幼鱼的存活率为60%,1 μg/mL的组中斑马鱼幼鱼的存活率为36%,2 μg/mL的组中斑马鱼幼鱼的存活率为15%,4 μg/mL组中的斑马鱼幼鱼的存活率为0%。结论:本实验硫酸铜对斑马鱼幼鱼具有致死的毒性且会影响其运动能力,中硫酸铜对斑马鱼的安全毒性范围是0.1~0.4 μg/mL,在安全毒性的范围之内进行炎症损伤的探索。本实验中72 h,0.4 μg/mL的组中斑马鱼全身性炎症损伤最明显。Objective: To complete the study, to measure the effects of different concentrations of copper sulfate on the developmental toxicity, and exercise ability of zebrafish and to elaborate the sensitivity of zebrafish to copper ion pressure in environmental water. Methods: Fish eggs were collected on day 0, pharmaceutical was dispensed on the third day, and dosing was started on the third day until the seventh day.
