摘 要:目的:借助网络药理学的方法探讨千金饮治疗慢性湿疹的作用机制。方法:通过CHEM-TCM、TCMID等数据库检索千金饮中药物含有的化合物,筛选其活性成分及其作用靶点;通过OMIM、DisGeNET和PubMed等数据库筛选与慢性湿疹相关的靶标,HPRD数据库进行PPI网络的构建,Cytoscape软件进行关键靶标的筛选,STRING数据库进行GO分析和信号通路富集分析。结果:共筛选出壮药千金饮的有效活性成分109个,对应273个作用靶点;慢性湿疹的相关基因靶标共有863个。千金饮治疗慢性湿疹的关键靶点包括NR3C1、NFKB1、YWHAE、HMGB1、PLG、TCF3、PPARG、VDR、TLR4、S100B等;GO功能富集分析主要涉及氮化合物代谢过程的负调控、细胞群增殖的调控、先天免疫反应、对外界刺激的反应调节等;信号通路富集分析主要涉及Toll样受体3 (TLR3)级联反应、TRIF (TICAM1)介导TLR4信号、TRAF6介导TLR7/8或9激活时NFkB和MAP激酶的诱导、细胞对应激的反应等信号通路。结论:千金饮可能通过调控机体的免疫、应激反应来治疗慢性湿疹,是多成分、多靶点、多途径相互作用的结果。Objective: To explore the mechanism of Qianjinyin in the treatment of chronic eczema with the help of network pharmacology. Methods: The compounds contained in Qianjinyin were searched by CHEM-TCM, TCMID database, and their active components and action targets were screened. OMIM, DisGeNET and PubMed databases were used to screen targets related to chronic eczema. PPI network was constructed in HPRD database, key targets were screened by Cytoscape software, and GO analysis and signal pathway enrichment analysis were performed in the STRING database. Results: A total of 109 active ingredients of Zhuangyao Qianjin Decoction were screened out, corresponding to 273 action targets. There were 863 gene targets associated with chronic eczema. Key targets of Qianjinyin in the treatment of chronic eczema include NR3C1, NFKB1, YWHAE, HMGB1, PLG, TCF3, PPARG, VDR, TLR4, S100B, etc. GO functional enrichment analysis mainly involves negative regulation of nitrogen metabolism, regulation of cell population proliferation, innate immune response, and regulation of response to external stimuli. Pathway enrichment analysis mainly involved Toll Like Receptor 3 (TLR3) Cascade, TRIF (TICAM1) mediated TLR4 signaling, TRAF6 mediated induction of NFkB and MAP kinases upon TLR7/8 or 9 activation, and cellular responses to stress. Conclusion: Qianjinyin may treat chronic eczema by regulating the immune and stress response of the body, which is the result of multi-component, multi-target and multi-pathway interaction.
