摘  要:目的观察钛(Ti)颗粒对RAW264.7巨噬细胞增殖及肿瘤坏死因子-α(TNF-α)、基质金属蛋白酶-9(MMP-9)表达的影响。方法 Ti颗粒设0、0.125、0.25、0.5和1 pg·mL^(-1)浓度组,观察RAW264.7巨噬细胞不同时段对各浓度Ti颗粒的吞噬作用;MTT法检测各浓度Ti刺激后RAW264.7巨噬细胞增殖能力;ELISA法检测0.125pg·mL^(-1)Ti刺激RAW264.7巨噬细胞前后分泌TNF-α、MMP-9的变化。结果 RAW264.7巨噬细胞出现吞噬Ti颗粒现象。MTT法检测表明所用各浓度Ti颗粒均不影响RAW264.7巨噬细胞增殖;RAW264.7巨噬细胞在0.125pg·mL^(-1) Ti颗粒的刺激下分别培养6、24和96h均可使NF-α、MMP-9表达增高(P<0.01)。结论 Ti颗粒可引起RAW264.7巨噬细胞吞噬反应,促进TNF-α、MMP-9表达,但不影响RAW264.7巨噬细胞的增殖。Objective To observe the vitality of RAW264.7 macrophages and the expression of TNF-α, MMP-9 by wear particles stimulating.Methodsthe morphological changes of RAW264.7 macrophages that titanium particles stimulated were observed for the different time duration. RAW264.7 macrophages viability was observed using the inverted microscope and the vitality of RAW264.7 macrophages was detected by titanium particles stimulated through methyl thiazolyl tetrazolium(MTT)colorimetric assay. Expression of TNF-αand MMP-9 were monitored by ELISA assay.Results Injecting titanium particles,RAW264.7 macrophages engulfed them with the increased concentration of titanium particles and RAW 264.7 macrophages proliferation declined. The cell viability of Titanium particles stimulating showed no significant difference, compared with control group. RAW264.7 macrophages vitality of titanium particles could cause RAW264. 7 macrophages phagocytosis reaction and promted by titanium particles(P<0.01). The expression of TNF-α,MMP-9 increased significantly by titanium particles stimulating for 6,24 and 96 hours(P<0.01). Conclusion RAW264.7 macrophage phagocytosis and cell viability were promted by wear particles stimulating. The expression of TNF-α,MMP-9 increased with wear particles stumulation while it did not affect cell viability.
