摘 要:目的:比较正常年轻组和正常老年组面部正常皮肤、皮肤鳞状细胞癌(cutaneous squamous cell carcinoma,cSCC)癌旁组织和cSCC组织中AKT、p-AKT、p53和Ki-67蛋白的表达情况,探讨PI3K-AKT信号转导通路在皮肤光老化及cSCC发生和发展中的相关机制。方法:蛋白质印迹法检测正常年轻组和正常老年组各12例面部正常皮肤组织中AKT和p-AKT的表达水平。免疫组织化学法检测正常年轻组和正常老年组各30例面部正常皮肤组织以及30例cSCC癌旁组织和30例cSCC组织中AKT、p-AKT、p53和Ki-67蛋白的表达水平。结果:蛋白质印迹法检测结果显示,正常年轻组与正常老年组面部正常皮肤组织中AKT和p-AKT蛋白的表达差异均无统计学意义(P>0.05)。免疫组织化学法检测结果显示,正常年轻组和正常老年组的正常面部皮肤组织以及cSCC癌旁组织和cSCC组织中AKT蛋白表达强度为(++/+++)的患者所占百分比分别为13.3%、20.0%、90.0%和93.3%,p-AKT蛋白表达强度为(++/+++)的患者所占百分比分别为6.7%、53.3%、86.7%和96.7%,p53蛋白表达强度为(+)的患者所占百分比分别为6.7%、53.3%、96.7%和96.7%,Ki-67蛋白表达强度为(+)的患者所占百分比分别为3.3%、6.7%、53.3%和60.0%。正常年轻组与正常老年组AKT蛋白的表达差异均无统计学意义(P>0.05),但是正常老年组p-AKT蛋白的表达水平显著高于正常年轻组(P<0.001)。cSCC癌旁组织和cSCC组织中AKT和p-AKT蛋白的表达水平均高于正常年轻组和正常老年组(P<0.001)。cSCC癌旁组织和cSCC组织中p53和Ki-67蛋白的表达水平均显著高于正常年轻组和正常老年组(P<0.001)。结论:PI3K-AKT信号转导通路可能在皮肤光老化和皮肤癌变过程中发挥作用,上调相关蛋白的表达。Objective:To compare the expressions of AKT,p-AKT,p53 and Ki-67 proteins in normal facial skin tissues of young patients and elderly patients and in the adjacent tissues of skin squamous cell carcinoma(cSCC)and cSCC tissues,and to explore the mechanism of PI3KAKT signal transduction pathway in skin photoaging and the occurrence and development of cSCC.Methods:The expression levels of AKT and p-AKT in normal facial skin tissues of 12 young patients and 12 elderly patients were detected by Western blotting.The expression levels of AKT,p-AKT,p53 and Ki-67 proteins in 30 normal facial skin tissues,30 adjacent tissues of cSCC and 30 tissues of cSCC were detected by immunohistochemistry.Results:The results of Western blotting showed that there was no significant difference in the expressions of AKT and p-AKT protein in normal facial skin tissues between the young patients and the elderly patients(P>0.05).The results of immunohistochemistry showed that the percentages of patients with AKT protein expression intensity of(++/+++)in normal facial skin tissues of young patients and the elderly patients,adjacent tissues of cSCC and cSCC tissues were 13.3%,20.0%,90.0%and 93.3%,respectively;the percentages of patients with p-AKT protein intensity of(++/++)were 6.7%,53.3%,86.7%and 96.7%,respectively;the percentages of patients with p53 protein expression intensity(+)were 6.7%,53.3%,96.7%and 96.7%,respectively;the percentage of patients with Ki-67 protein expression intensity(+)were 3.3%,6.7%,53.3%and 60.0%,respectively.There was no significant difference in AKT protein expression between young patients and elderly patients(P>0.05),but the expression level of p-AKT protein in elderly patients was significantly higher than that in young patients(P<0.001).The expression levels of AKT and p-AKT protein in the adjacent tissues and cSCC tissues were higher than those in the young patients and elderly patients(P<0.001).The expression levels of p53 and Ki-67 protein in the adjacent tissues of cSCC and cSCC tissues were significantly hi
