摘　　要：多腺苷二磷酸核糖聚合酶[poly(ADP-ribose)polymerase,PARP]在DNA损伤修复过程中发挥重要作用。近年来,有关PARP与肿瘤发生的关系及PARP抑制剂在联合治疗中通过“合成致死”效应增强抗肿瘤疗效的研究取得了显著成果。已报道的与PARP抑制剂联合使用的治疗方式包括放疗、化疗、免疫检查点抑制剂、血管生成抑制剂及其他靶向药物治疗。本文就PARP抑制剂与这些抗肿瘤治疗手段联用在恶性肿瘤治疗中的作用机制、疗效及影响因素的研究进展进行了综述。Poly(ADP-ribose)polymerase(PARP)plays significant roles in DNA damage repair.Remarkable achievements have been accomplished in the studies of the relationship between PARP and oncogenesis,as well as the enhancement of therapeutic effect through PARP-inhibitorinduced“synthetic lethality”in combined therapies.PARP inhibitors have been reported to be used in combination with radiotherapy,chemotherapy,immune checkpoint inhibitors,angiogenesis inhibitors and other targeted drugs in cancer therapy.This review discussed about the recent research progress on the mechanism,efficacy and influencing factors of PARP-inhibitor-combined therapies in cancer treatment.
