摘  要:目的探究α_(1)肾上腺素能受体(α_(1)AR)各亚型在肝窦内皮细胞(LSEC)中的表达情况。方法体外培养人原代LSEC,蛋白质印迹法验证α_(1)AR激动剂去氧肾上腺素和α_(1)AR阻滞剂多沙唑嗪对LSEC表达血管生成相关指标CD31和vWF的影响,并验证LSEC中α_(1)AR各亚型的表达情况。免疫组化染色验证小鼠肝脏α_(1)AR各亚型的表达情况。结果α_(1)AR激动剂去氧肾上腺素可增加LSEC中CD31和vWF的表达,而α_(1)AR阻滞剂多沙唑嗪可拮抗此作用。体内和体外实验均在LSEC中检测到α_(1a)AR与α_(1b)AR的表达,未检测到α_(1d)AR的表达。结论α_(1)AR参与LSEC表型的调控,且通过α_(1a)AR与α_(1b)AR两亚型实现此功能,因此α_(1)AR可能成为肝纤维化新的调控靶点。Objective To investigate the expressions ofα_(1)adrenergic receptor(α_(1)AR)subtypes in liver sinusoidal endothelial cells(LSEC).Methods Human primary LSEC were cultured in vitro.The effects ofα_(1)AR agonists phenylephrine andα_(1)AR blocker doxazosin on the expressions of angiogenesis-related markers CD31 and vWF in LSEC and the expressions of variousα_(1)AR subtypes in LSEC were verified by Western blotting analysis.Immunohistochemistry was used to verify the expressions ofα_(1)AR subtypes in mouse liver.Resultsα_(1)AR agonist phenylephrine could increase the expressions of CD31 and vWF in LSEC,andα_(1)AR blocker doxazosin antagonized this effect.The expressions ofα_(1a)AR andα_(1b)AR were detected in LSEC in vivo and in vitro,but the expression ofα_(1d)AR was not detected.Conclusionsα_(1)AR is involved in the regulation of LSEC phenotype,and this function is achieved through two subtypes ofα_(1a)AR andα_(1b)AR.Therefore,α_(1)AR may become a new regulatory target of liver fibrosis.
