摘 要:目的探讨人胎盘羊膜来源的间充质干细胞(h AMSCs)向乳腺肿瘤相关成纤维细胞(CAFs)转化后,对乳腺癌细胞上皮间质转化(EMT)的影响。方法选取2017年7月至2018年7月间牡丹江医学院附属红旗医院收住的80例产妇的足月剖腹产胎儿羊膜。MCF-7单独培养或Transwell小室共培养人乳腺癌细胞系MCF-7和MSCs共培养组即试验组,MCF-7细胞MSC培养基组为对照组,MCF-7常规培养组为对照组2。采用RT-qPCR检测共培养后h AM-MSCs向CAFs转化及CAFs相关特征标志物平滑肌肌动蛋白-α(α-SMA)和腱生蛋白-c(tenascin-c)的基因表达。对共培养后的肿瘤细胞进行细胞EMT相关标志及肿瘤特性检测。结果与对照组比较,共培养试验组MCF-7细胞发生上皮间质转化。与MSCs共培养能明显促进乳腺癌细胞的侵袭和迁移。结论人胎盘羊膜来源的间充质干细胞可诱导乳腺癌细胞上皮间质转化。Objective To study the effect of human placental amnion-derived mesenchymal stem cells(h AM-MSCs)after transformation into tumor-associated fibroblasts(CAFs)on epithelial-mesenchymal transition(EMT).Methods Human placental amnion was collected from 80 women who gave a full-term cesarean birth at Hongqi Hospital Affiliated to Mudanjiang Medical College between July 2017 and July2018.Breast cancer cell line(MCF-7)was cultured alone or indirectly co-cultured with h AM-MSCs using Transwell system.The co-cultured was included in a study group.MCF-7 cultured on the media of MSC was included in control group 1 and MCF-7 cultured conventionally was included in control group 2.RT-qPCR was used to detect the transformation of co-cultured MCF-7 into CAFs and the expression of markers of CAFs-related markersα-smooth muscle actin(α-SMA)and tenascin-C.EMS testing was performed in the co-cultured MCF-7 cells for the relevant markers as well as characteristic of cancer were analyzed.Results Following indirect co-culture with h AM-MSCs,EMT concurred in MCF-7 and co-culture with h AM-MSCs can promote cell invasion and migration.Conclusion h AD-MSCs can stimulate epithelial-mesenchymal transition of breast cancer cells.
