摘 要:目的探讨透明质酸联合A型肉毒毒素(botulinum toxin type-A,Botox A)和富含血小板血浆(platelet rich plasma,PRP)进行猪真皮内注射,检测真皮层成纤维细胞数目及Ⅰ型胶原蛋白mRNA分泌代谢的变化。方法采用巴马猪建立动物模型,使用交联透明质酸(cross-linked hyaluronic acid,HA)和非交联透明质酸(non-cross-linked hyaluronic acid,N-HA)分别与Botox A、PRP联合作为实验组(HA组、HA+Botox A组、HA+PRP组、N-HA组、N-HA+Botox A组、N-HA+PRP组),并与生理盐水组和正常组比较。于注射第1和4周后,通过组织切片染色、实时荧光定量聚合酶链反应(PCR)等方法,定性定量观察、比较真皮层成纤维细胞数量变化,真皮层Ⅰ型胶原蛋白mRNA分泌表达变化。结果HA组的成纤维细胞数量、Ⅰ型胶原蛋白mRNA表达量在第1周和第4周时比N-HA组高,并且HA+PRP组的成纤维细胞数量、Ⅰ型胶原蛋白mRNA表达水平显著高于同期HA+Botox A组、生理盐水组和正常组(P<0.05)。结论应用HA与PRP联合注射更易促进成纤维细胞的增殖和Ⅰ型胶原蛋白mRNA的表达,其主要机制可能是通过改变细胞外基质的理化性质进而促进真皮层细胞因子的合成,从而影响胶原合成。Objective The aim of this study is to investigate the effects of hyaluronic acid combined with botulinum toxin type A(Botox A)and platelet-rich plasma(PRP)on the secretion of collagen-Ⅰin the porcine dermis.Methods The animal model was established in bama pigs.Cross-linked hyaluronic acid(HA)and non-cross-linked hyaluronic acid(N-HA)were used as experimental groups in combination with Botox A and PRP,respectively:HA,HA+Botox A,HA+PRP,N-HA,N-HA+Botox A,N-HA+PRP.Those were compared to control groups,one positive control group:saline group and one negative control group:control group without any intervention.As variables,the number of dermal fibroblasts,the expression of collagen-ⅠmRNA were measured by real-time fluorescence quantitative PCR,as well as by qualitative tissue staining 1 and 4 weeks post injection.Results The expression of fibroblasts and collagen-ⅠmRNA in the HA group were higher than those in the N-HA group.At the same time,the number of fibroblasts,and the expression of collagen-ⅠmRNA in HA+PRP group were significantly higher than those in HA+Botox A group,saline group and control group(P<0.05).Conclusion Application of HA combined with PRP is more likely to promote fibroblast proliferation and collagen-ⅠmRNA expression.The main mechanism may be by changing of physical and chemical properties of the extracellular matrix and promoting dermal cytokine synthesis as well as stimulating the collagen mRNA expression.
