血管紧张素在脑室周器官及丘脑下部的分布及其对饮水行为的神经内分泌控制

摘  要:本文以电破坏穹窿下器官,以霍乱毒素—辣根过氧化酶追综及[[^(14)C]]-脱氧葡萄糖放射自显影电子计算机图象程序处理等综合技术,观察Sprague~Dawley系大鼠在脑室及静脉内注入血管紧张素Ⅱ后,脑室周器官及丘脑下部的行为神经内分泌作用。结果表明:穹窿下器官、正中隆起、弓状核、视上核、室旁核及神经垂体的[[^(14)C]]-脱氧葡萄糖摄取相对视密度明显增高,提示这些结构在血管紧张素致渴饮水行为中,调节水盐平衡的神经内分泌作用。穹窿下器官是感受血管紧张素最敏感结构之一,它与丘脑下部的神经内分泌核群密切联系。电破坏穹窿下器官则大鼠失去在迷宫内快速奔跑饮水行为。Intracerebroventricular injection of angiotensinⅡelicits drinking,vasopressin release and pressor responses.The possible central sites of angiotensin action are the circumventricular organ and the bypothalamus.Lesioning the Subfornical organ(SFO)eliminated the drinking behavior in the runway after physiologically meaningful doses of angiotensinⅡgiven intravenously.Labelled terminals were found in nucleus medianus of the medial preoptic area(NM),organum vasculosum of the lamina terminalis(OVLT),supraoptic nuclei(SON)and paraventricular nuclei(PVN)following the injection of CT-HRP into SFO.The[^(14)C]-2DG functional autoradiography technique and computerized image-processing revealed changes in Relative Optical Density between conscious Sprague-Dawley rats either receiving a pulse intracerebroventricular injection of angiotensinⅡ(60ng)or no injection(control).The results demonstrated significant increases in activity in the cireumventricular organs:SFO,median eminence(ME),and in the hypothalamic structures SON,PVN,arcuate nuclei(ARC)and posterior pituitary.Other structures observed were not significantly affected by the treatment with angiotensinⅡ.We concluded that intracranial angiotensinⅡis a stimulant of metabolic activity in certain cireumventricular organs and hypothalamic structures.And these structures may have neuroendocrinic influence on the behavior of drinking.
