摘  要:体外三种胃癌细胞株的染料排斥试验及~3H—胸腺嘧啶掺入试验均证实石蒜碱内铵盐(AT-1840)及细小病毒H-1(PVH-1)对胃癌细胞具有直接杀伤作用,药物浓度与杀伤强度呈正比,最低有效浓度分别为0.1μg/孔和1×10~4/孔。流式细胞仪检测证实AT-2840作用于G2/M增殖期胃癌细胞。PVH-1作用于S期胃癌细胞。PVH-1感染肿瘤细胞后即掺入细胞核内增殖,细胞膜绒毛变性、丢失,其后细胞肿胀、崩解。两药最低有效剂量联用,对胃癌细胞杀伤作用起协同增强效应,抗肿瘤作用可超过任一单独应用最大剂量时的作用强度。AT-1840使实体瘤重量明显减少,PVH-1和AT-1840明显延长带胃癌裸小鼠(腹水型及实体型)的生存期。本研究首次证实AT-1840和PVH-1均为低浓度有效的胃癌细胞直接杀伤因子,且具有协同增强作用。This study tried to demonstrate the in vitro killing effect of AT-1840(a Chinese herbal extract)on gastric cancer cells and also to identify the sites of action on cell cycle of these cells.The dye exclusion test and~3H-thymidine incorporation have demonstrated that AT-1840 and parvovirus H-1(PVH-1)have direct cytocidal effect on stomach cancer cells.Combined use of AT-1840 and PVH-1 at the lowest concentrations increased the.cytocidal effect markedly.Using flow cytometry,it was found that both drugs were cell cycle specific agents,there was high accumulation of G/M phase cells for AT-1840 and S phase cells for PVH-1.With transmission and scanning electron mic(?)oscopy,it was found that once cancer cells were infected with PVH-1,this virus could penetrate into the nuclei of cancer cells and replicated,and resulted in the shortening of cancer cells villi and cell lysis ensued.In stomach cancer-bearing nude mice,both A T-1840 and PV H-1 prolonged the survival period and decreased the size of solid tumors.
