摘 要:将指数生长期的球等鞭金藻(Isochrysisg albana)H24和棕鞭藻(Ochrosphaero neopolitana)H32接种到1000 m L三角烧瓶的2f培养基中,第8 d添加0 mmol/L(对照组)、0.5 mmol/L、1.5 mmol/L、2.5 mmol/L和5.0mmol/L的钨酸钠(Na_(2)WO_(4)),光强为100μmol·m^(-2)s^(-1),连续光照、充气,水温25℃,盐度31下培养5 d,探讨不同浓度Na_(2)WO_(4)对2株海洋金藻的生长及脂质积累的影响。结果表明,Na_(2)WO_(4)浓度对2株金藻的细胞密度、叶绿素含量、叶绿素荧光参数Fv/Fm(光系统II最大光化学效率)、r ETR(相对电子传递速率)、q P(光化学猝灭)和脂肪酸组成均有显著影响(P<0.05)。Na_(2)WO_(4)浓度对球等鞭金藻的总脂含量有显著影响(P<0.05)。0 mmol/L及0.5mmol/L Na_(2)WO_(4)浓度组2株金藻的最终细胞密度最大。在2.5 mmol/L Na_(2)WO_(4)浓度组,球等鞭金藻Fv/Fm值最高(第4~5 d),在0 mmol/L和2.5 mmol/L Na_(2)WO_(4)浓度组棕鞭藻的Fv/Fm值最高(第1~2 d)。2种金藻的总脂含量随Na_(2)WO_(4)浓度升高而增加,在5.0mmol/L时达到最大值(分别为31.91%和25.55%),分别比对照组高38.21%和25.24%。在5.0 mmol/L Na_(2)WO_(4)时,球等鞭金藻的18∶1(n-9)(34.54%)、饱和脂肪酸(SFA)(37.59%)和单不饱和脂肪酸(MUFA)(42.65%)含量达最大值;对照组多不饱和脂肪酸(PUFA)含量最高(36.23%),之后随Na_(2)WO_(4)浓度增高呈下降趋势。棕鞭藻的16∶0(14.69%)和SFA(32.15%)含量在5 mmol/L达最大值,对照组18∶1(n-9)含量最高(37.33%);二十碳五烯酸(EPA,20∶5(n-3))和PUFA含量随Na_(2)WO_(4)浓度升高先增加后降低,2.5 mmol/L时达最大值,分别为10.72%和31.96%。Two species in Chrysophyta Isochrysisg albana H24 and Ochrosphaero neopolitana H32 in the exponential growth phase were inoculated and cultured into 2f culture medium in a 1000 m L Erlenmeyer flask,and sodium tungstate dihydrate(Na_(2)WO_(4))was added into the flask at a dose of 0 mmol/L(control group),0.5 mmol/L,1.5 mmol/L,2.5 mmol/L and 5.0 mmol/L on the 8th day at light intensity of 100μmol·m^(-2)s^(-1),continuous light and aeration,at water temperature of 25℃,and a salinity of 31 for five days to investigate the effect of different Na_(2)WO_(4)concentrations on the growth and lipid accumulation of the two Chrysophyta algae.The results showed that the cell density,chlorophyll content,chlorophyll fluorescence parameters(Fv/Fm,maximal photochemical efficiency in PSII),r ETR(relative electron transport rate),q P(photochemical quenching)and fatty acid compositions of two Chrysophyta algae were significantly influenced by Na_(2)WO_(4)concentrations(P<0.05)and the total lipid content of I.galbana was significantly influenced by Na_(2)WO_(4)concentrations(P<0.05),with the maximal final cell densities of the two Chrysophyta algae in mmol/L)and the low concentration of Na_(2)WO_(4)(0.5 mmol/L)group.The maximal Fv/Fm value was found in I.galbana at middle concentration of Na_(2)WO_(4)(2.5 mmol/L)(4~5 d)and the maximal Fv/Fm value in O.neopolitana was observed at 0 mmol/L and 2.5mmol/L Na_(2)WO_(4)(1~2 d).The total lipid contents in the two Chrysophyta algae were shown to be increased with the increase in concentration of Na_(2)WO_(4),with the maximum 31.91%and 25.55%at 5.0 mmol/L Na_(2)WO_(4),and 38.21%and 25.24%higher than those at 0 mmol/L Na_(2)WO_(4),respectively.The maximal contents of 18∶1(n-9)(34.54%),saturated fatty acid(SFA)(37.59%)and mono unsaturated fatty acid(MUFA)(42.65%)in I.galbana were found at 5.0 mmol/L Na_(2)WO_(4).The contents of PUFA(polyunsaturated fatty acid)in I.galbana were decreased with the in crease in concentration of Na_(2)WO_(4),and the maximum value(36.23%)appeared in control group
