摘　　要：目的研究LncRNA人类白细胞抗原复合体18(HCG18)和miR-605-3p在脑胶质瘤中的表达及其临床意义。方法选取2017年8月-2018年8月在沧州市中心医院行手术治疗的脑胶质瘤患者112例为脑胶质瘤组,取其手术切除的脑胶质瘤组织作为研究标本;另选取同期行颅脑损伤内减压术的患者114例作为对照组,取其正常脑组织作为研究标本。qRT-PCR法测定两组脑组织中LncRNA HCG18和miR-605-3p表达水平,Pearson法分析脑胶质瘤组织中LncRNA HCG18和miR-605-3p表达相关性。收集脑胶质瘤患者临床病理特征,分析脑胶质瘤组织中LncRNA HCG18和miR-605-3p表达水平(以LncRNA HCG18和miR-605-3p表达水平的平均数为界限,分为高、低表达组)与患者临床病理特征的关系。对脑胶质瘤患者术后进行随访,Kaplan-Meier法分析脑胶质瘤组织中LncRNA HCG18和miR-605-3p表达水平与患者预后的关系。结果与对照组比较,脑胶质瘤组LncRNA HCG18表达水平升高(1.01±0.19)vs.(2.32±0.31),miR-605-3p表达水平降低(1.02±0.17)vs.(0.31±0.08),差异有统计学意义(t=38.376、40.055,P均<0.05)。Starbase网站预测LncRNA HCG18和miR-605-3p存在靶向结合位点。相关性分析显示,脑胶质瘤组织中LncRNA HCG18与miR-605-3p成负相关(r=-0.756,P<0.05)。与肿瘤分级Ⅰ~Ⅱ级比较,Ⅲ~Ⅳ级脑胶质瘤组织中LncRNA HCG18高表达占比较高,miR-605-3p低表达占比较高,差异有统计学意义(P<0.05)。Kaplan-Meier分析结果显示,LncRNA HCG18高表达组累积生存率(47.7%)低于LncRNA HCG18低表达组(61.1%),差异有统计学意义(χ^(2)=6.038,P=0.014);miR-605-3p低表达组累积生存率(37.7%)低于miR-605-3p高表达组(69.1%),差异有统计学意义(χ^(2)=13.055,P<0.001)。结论LncRNA HCG18和miR-605-3p均在脑胶质瘤组织中异常表达,二者成负相关,均与脑胶质瘤患者的疾病进展以及预后相关。Objective To study the expression and correlation of LncRNA human leukocyte antigen complex 18(HCG18)and miR-605-3p in glioma.Methods A total of 112 patients with glioma who underwent surgical treatment in our hospital from August 2017 to August 2018 were included in this study,and the surgically resected glioma tissue was taken as the research specimen.Another 114 patients who underwent internal decompression of craniocerebral injury in our hospital were regarded as the control group,and the normal brain tissue was taken as the research specimen.The expression levels of LncRNA HCG18 and miR-605-3p in the two groups were determined by qRT-PCR;the correlation between the expressions of LncRNA HCG18 and miR-605-3p in glioma tissue was analyzed by Pearson method;the clinicopathological characteristics of glioma patients were collected,and the relationship between the expression levels of LncRNA HCG18 and miR-605-3p in glioma tissue and the clinicopathological characteristics of patients was analyzed(based on the average of the expression levels of LncRNA HCG18 and miR-605-3p,they were divided into high and low expression groups).Patients with glioma were followed up after operation,and the Kaplan-Meier method was performed to analyze the relationship between the expression levels of LncRNA HCG18 and miR-605-3p in glioma tissue and the prognosis of patients.Results Compared with the control group,the expression level of LncRNA HCG18 in brain glioma tissue was increased(1.01±0.19)vs.(2.32±0.31),and the expression level of miR-605-3p was reduced(1.02±0.17)vs.(0.31±0.08)(t=38.376,40.055,all P<0.05).The Starbase predicted that there were targeted binding sites for LncRNA HCG18 and miR-605-3p;correlation analysis showed that LncRNA HCG18 was negatively correlated with miR-605-3p in glioma tissue(r=-0.756,P<0.05).Compared with tumor gradesⅠ-Ⅱ,the proportion of high expression of LncRNA HCG18 in gradesⅢ-Ⅳglioma tissues was higher(P<0.05);compared with tumor gradesⅠ-Ⅱ,the proportion of low expression of miR-6
