摘 要:目的探讨富含亮氨酸的重复序列蛋白(leucine-rich repeat-containing protein 31,LRRC31)在前列腺癌(prostate cancer,PCa)患者中的表达水平及对临床预后意义。方法通过在肿瘤基因图谱(the cancer genome atlas,TCGA)数据库下载并分析前列腺相关患者基因表达数据共500例,其中前列腺癌数据490例,导管及小叶肿瘤9例,囊性、黏液性及浆液性肿瘤1例。明确LRRC31在前列腺癌组织与正常前列腺组织之间的表达差异,分析LRRC31的表达与前列腺癌患者年龄、分期以及总生存时间的相关性,探讨LRRC31对于前列腺癌细胞的作用机制。结果LRRC31在前列腺癌细胞中的表达水平明显高于其在癌旁组织中的表达水平,差异有统计学意义(P=0.022),LRRC31的表达在患者年龄分布上差异无统计学意义(P=0.347);高表达LRRC31的前列腺癌患者的T分期明显高于低表达的患者,差异有统计学意义(P<0.001);发生周围淋巴结受累的前列腺癌患者的LRRC31的表达水平与未发生周围淋巴结受累的患者的LRRC31的表达水平比较,差异无统计学意义(P=0.123);LRRC31低表达的前列腺癌患者的总生存期明显优于LRRC31高表达的前列腺癌患者,差异有统计学意义(P=0.024)。根据基因集富集分析(gene set enrichment analysis,GSEA)结果表明,LRRC31可能通过多种细胞代谢途径及细胞通路影响前列腺癌的发生,包括谷胱甘肽代谢、Notch信号通路、苯丙氨酸代谢、RIG-I样信号通路、RNA降解、小细胞肺癌及泛素介导的蛋白水解途径。结论LRRC31基因与前列腺癌患者的不良预后相关,可以作为前列腺癌的独立危险因素。Objective This study aimed to explore the expressionlevel of leucine-rich repeat-containing protein 31(LRRC31)in patients with prostate cancer(PCa),and its clinical prognostic significance.Method PCa gene expression data from The Cancer GenomeAtlas(TCGA)database were downloaded and analyzed,including a total of 500 patients with prostate-related gene expression data.Among them,490 cases were prostate cancer,9 cases were ductal and lobular tumors,and 1 case was a cystic,mucinous,or serous tumor.First,we compared the differences in LRRC31 expression between PCa and normal prostate tissue.We determined the correlation between LRRC31 expression and the clinical features of patients with PCa,such as age,T stage,N stage and over all survival.Finally,we investigated the potentially relevant mechanism of LRRC31 on PCa cells.Result LRRC31 expression was significantly higher in PCa than paracancer tissue(P=0.022).No significant difference was observed in the age distribution according to LRRC31 expression(P=0.347).Patients with PCa with high LRRC31 expression had higher T staging than those with low LRRC31 expression.No significant difference was observed in LRRC31 expression in patients with PCa with versus without peripheral lymph node involvement(P=0.123).Overall survival was prolonged in patients with lower than higher expression of LRRC31(P=0.024).According to the results of GSEA enrichment analysis,LRRC31 may promote the growth of PCa cells through several biological processes and signaling pathways,including glutathione metabolism,the Notch signaling pathway,phenylalanine metabolism,the RIGI like receptor signaling pathway,RNA degradation,small cell lung cancer,and ubiquitin mediated proteolysis.Conclusion The LRRC31 gene is associated with poor prognosis in PCa patients and might be an independent risk factor for PCa.
