摘  要:目的探讨腺苷预处理对大鼠脑缺血再灌注后血脑屏障(BBB)通透性和紧密连接蛋白occludin表达的影响。方法采用大鼠大脑中动脉栓塞(MCAO)模型,伊文思蓝(EB)渗透性实验检测BBB通透性,2,3,5-氯化三苯基四氮唑染色(TTC)计算脑梗死体积百分比,免疫组织化学法测定occludin蛋白表达,RT-PCR法测定occludin mRNA表达。结果与假手术组比较,模型组、腺苷组大鼠脑组织中EB含量均明显升高,脑梗死体积显著增加,occludin蛋白及mRNA表达均显著减少(P<0.01);与模型组比较,腺苷组大鼠脑组织中EB含量显著降低,脑梗死体积显著减小,脑组织中occludin蛋白及mRNA表达明显升高(P<0.01)。结论腺苷通过上调脑缺血再灌注后脑组织中occludin的表达降低BBB通透性,减小脑梗死体积。Objective To study the effects of adenosine pretreatment on blood brain barrier(BBB)and occludin expression after cerebral ischemia-reperfusion in rats.Methods Using middle cerebral artery occlusion(MCAO)model in rats,and the BBB permeability was assessed by Evans blue extravasation.The percentage of cerebral infraction area was calculated by 2,3,5-TTC staining.The expression of occludin protein and mRNA were determined by immunohistochemistry assay and reverse transcriptase-polymerase chain reaction,respectively.Results Compared with sham group,EB content was increased significantly,the percentage of cerebral infraction area was increased significantly,the expression of occludin protein and mRNA was decreased markedly in brain tissue of model group and adenosine group(P<0.01).Compared with model group,EB content was markedly decreased,the percentage of cerebral infraction area was markedly decreased,the expression of occludin protein and mRNA was increased markedly in adenosine group(P<0.01).Conclusion Adenosine reduces BBB permeability and cerebellar infarct volume by up-regulating occludin expression in cerebral tissue after cerebral ischemia reperfusion.
