摘　　要：目的探索ARMC7在膀胱癌细胞中的表达情况及其对膀胱癌细胞增殖和迁移能力的影响。方法下载Human Protein Atlas数据库中膀胱尿路上皮癌的生存预后数据,并应用R软件中的survminer包选择ARMC7表达的最佳截止值进行生存分析;采用Western blot方法检测膀胱癌细胞系以及人正常膀胱上皮细胞SVHUV-1中ARMC7的蛋白表达情况;构建稳定高表达ARMC7的J82和5637细胞系;采用CCK-8检测转染ARMC7对膀胱癌细胞J82和5637增殖能力的影响;采用划痕实验检测转染ARMC7对膀胱癌细胞J82和5637迁移能力的影响。结果膀胱癌组织中ARMC7的低表达与膀胱癌患者的不良预后显著相关(P<0.05);膀胱癌细胞系中ARMC7的蛋白表达显著低于人正常膀胱上皮细胞SV-HUV-1(P<0.01),ARMC7高表达能够显著抑制膀胱癌细胞的增殖和迁移能力(P<0.01)。结论ARMC7能够抑制膀胱癌细胞的增殖和迁移,且其表达水平与膀胱癌患者的预后显著相关。Objective To explore the expression of ARMC7 in bladder cancer cells and its effect on the proliferation and migration of bladder cancer cells.Methods The survival data of bladder urothelial cancer was downloaded from the Human Protein Atlas database,and then the survival analysis is performed by using the R survminer package to select the best cutoff value for ARMC7 expression.Western blot was used to detect ARMC7 protein expression levels in bladder cancer cell lines and human normal bladder epithelial cells SV-HUV-1.Stable transfection technology is used to construct stable and highly ARMC7-expressing bladder cancer cell J82 and 5637.CCK-8 assay is used to detect the effect of high expression of ARMC7 on the proliferation of bladder cancer cells J82 and 5637.Wound-healing assay was used to detect the effect of high expression of ARMC7 on the migration of bladder cancer cells J82 and 5637.Results The low expression of ARMC7 in bladder cancer tissue was significantly related to the poor prognosis of patients with bladder cancer(P<0.05).The protein expression of ARMC7 in bladder cancer cell lines was significantly lower than that of human normal bladder epithelial cells SV-HUV-1(P<0.01).The high expression of ARMC7 in bladder cancer cells can significantly inhibit the proliferation and migration of bladder cancer cells(P<0.01).Conclusion ARMC7 can inhibit the proliferation and migration of bladder cancer cells,and its expression level is significantly related to the prognosis of bladder cancer patients.
