摘  要:本研究旨在探究水牛(Bubalus bubalis)载脂蛋白B基因(ApoB)在地中海水牛群体中的遗传多态性,并与地中海水牛泌乳性状进行关联分析,筛选水牛产奶性状分子标记。本研究以350头地中海纯种奶水牛为研究对象,利用直接测序法和基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)法筛选基因多态性(single nucleotide polymorphisms,SNPs)与基因分型,并进一步分析单倍型与泌乳性状的相关性。结果显示,在地中海水牛群体ApoB基因中筛选到10个SNPs,分别位于启动子(g.-1718A>G、g.-1823G>C和g.-2063A>C)、第7内含子(g.8105G>A)、第26内含子(g.30643G>A)、第5外显子(g.5903G>A)、第26外显子(g.30879T>C、g.33638A>C和g.37375G>A)和第29外显子(g.40787A>G)区域。10个SNPs位点多态信息含量均大于0.25且小于0.5,属于中度多态。其中g.-1718A>G、g.-1823G>C、g.-2063A>C、g.37375G>A和g.40787A>G突变位点与地中海水牛乳蛋白率显著相关,g.5903G>A和g.40787A>G位点与地中海水牛乳脂率显著相关,g.37375G>A与地中海水牛270天乳蛋白量和乳脂量显著相关。单倍型Block1(g.-1718A>G、g.-1823G>C和g.-2063A>C)与270天乳蛋白量和乳蛋白率显著相关;单倍型Block2(g.30643G>A和g.33638A>C)与270天乳脂含量、乳脂率、270天乳蛋白量以及乳蛋白率显著相关;单倍型Block3(g.37375G>A和g.40787A>G)与乳蛋白率显著相关;g.33638A>C、g.37375G>A、g.40787A>G以及单倍型Block2和Block3突变型蛋白质理化性质和二级结构发生改变。本研究共筛选到6个SNPs和3个单倍型Block与水牛泌乳性状显著相关,其中单倍型Block2的H3H3为最优单倍型,可作为水牛产奶性状潜在分子标记。This study aims to identify ApoB polymorphisms in buffalo(Bubalus bubalis),and to further explore the molecular markers associated with buffalo lactation traits.In the present study,ApoB single nucleotide polymorphisms(SNPs)were identified by DNA sequencing in a total of 350 Mediterranean buffaloes,and MALDITOF-MS was employed for genotyping.Association analysis between ApoB haplotype and milk production traits was carried by using SAS9.4 software.Besides,the physical,chemical properties and structure of ApoB protein were predicted by using bioinformatic tools.In the present study,10 SNPs were screened,3 of which located in the promoter(g.-1718A>G,g.-1823G>C,and g.-2063A>C),intron 7(g.8105G>A),1 of which located in the intron 26(g.30643G>A),1 of which located in the exon 5(g.5903G>A),3 of which located in the exon 26(g.30879T>C,g.33638A>C,g.37375G>A)and 1 of which located in the exon 29(g.40787A>G).All the identified SNPs were moderately polymorphic(0.25<PIC<0.50).Association analysis showed that g.-1718A>G,g.-1823G>C,g.-2063A>C,g.37375G>A,and g.40787A>G were significantly related to milk protein percentage,g.5903G>A and g.40787A>G were significantly related to milk fat percentage,and g.37375G>A was significantly related to 270-day milk protein and 270-days milk fat yield.Haplotype block1(g.-1718A>G,g.-1823G>C,and g.-2063A>C)was significantly associated with 270-days milk protein yield and milk protein percentage.Haplotype block2(g.30643G>A and g.33638A>C)was significantly related to 270-days milk fat and milk protein yield,milk fat percentage,and milk protein percentage.Haplotype block3(g.37375G>A and g.40787A>G)was significantly related to the milk protein percentage,g.33638A>C,g.37375G>A,g.40787A>G and haplotype block3(g.37375G>A and g.40787A>G)affected the physical and chemical properties and the secondary structure of the ApoB protein.In conclusion,a total of 6 SNPs and 3 haplotype blocks were identified significantly associated with buffalo milk production traits.Among them,H3H3 in haplotype block2 was the
