摘 要:目的建立分光光度法检测肉制品中亚硝酸盐含量的方法.方法准确称取经粉碎匀浆的样品,采用饱和硼砂提取,沉淀蛋白质及离心除去脂肪,过滤后加入重氮化试剂及偶合试剂进行相关反应显色,测量吸光度,外标法定量.以CFAPA-1387《肉制品中硝酸盐,亚硝酸盐的测定能力验证计划》进行方法验证.结果亚硝酸盐在0.02~0.40μg/mL质量浓度范围内具有良好的线性关系,相关系数在0.999以上,平均回收率为98.9%~101.3%,相对标准偏差为0.36%~0.87%.CFAPA-1387能力验证检测值分别为3.25 mg/kg和1.85 mg/kg,对应判定Z比分值为-0.47和-0.12,考核结果为满意.结论优化的检测方法具有较好的重复性和准确率,样品前处理操作简便,结果可靠,适合卫生应急事件的快速处置及常规实验室进行肉制品亚硝酸盐的检测.Objective To establish a spectrophotometric method for the determination of nitrite in meat products.Methods The samples were accurately weighed and crushed homogenized,extracted with saturated borax,precipitated protein,centrifuged to remove fat.After filtration,diazotization reagent and coupling reagent were added for related reaction color development,absorbance was measured and quantitative by external standard method.The method was verified by CFAPA-1387 Verification plan for the determination of nitrate and nitrite in meat products.Results Nitrite had a good linear relationship in the concentration range of 0.02-0.40μg/mL,the correlation coefficient was above 0.999,the average recoveries were 98.9%-101.3%,and the relative standard deviations were 0.36%-0.87%.The ability verification test values of CFAPA-1387 were 3.25 mg/kg and 1.85 mg/kg,respectively,and the corresponding Z-score values of the decision were-0.47 and-0.12,and the assessment results were satisfactory.Conclusion The optimized detection method has good repeatability and accuracy,simple sample pretreatment operation,reliable results,suitable for the rapid disposal of health emergency and routine laboratory detection of nitrite in meat products.
