槲皮素对顺铂诱导的结肠癌HT-29细胞凋亡增敏作用的研究

摘　　要：目的探讨槲皮素对顺铂诱导结肠癌HT-29细胞生长抑制、诱导凋亡及对NF-κB表达的影响.方法采用MTT法、流式细胞仪,以及现在常用的免疫组化新技术,用来致力于槲皮素的研究,将顺铂,槲皮素以及顺铂三种药物相互结合,进而得到的新药物,可以有效的作用于肝癌细胞,进而很好的对其后生长进行抑制、形态学变化、以及相应的诱导凋亡和对NF-κB表达的影响.结果MTT法证实联合,可以有效的抑制相应的作用,时常满24 h、48 h后(A 0.489±0.020→0.298±0.096,0.799±0.043→0.311±0.018,P<0.01).流式细胞仪对应的检测性质最强,细胞周期阻滞于G2/M期.免疫组化技术检测顺铂作用后NF-κB表达(63.8±5.6)%,联合组作用后(23.2±2.3)%(P<0.01).结论槲皮素增强顺铂诱导结肠癌HT-29细胞凋亡,其机制可能是通过抑制激活NF-κB的表达,提示槲皮素与顺铂联合有望应用于肿瘤联合化疗.Objective To investigate effect of quercetin on growth inhibition,induction of apoptosis and NF-κB expression of cisplatin-induced colon cancer HT-29 cell.Methods With MTT method,flow cytometry,and commonly used new technology of immunohistochemistry,the article carried on research of quercetin,combined cisplatin,quercetin and cisplatin with each other to obtain new drugs,which could act on liver cancer cells effectively,affect their subsequent growth inhibition,morphological changes,and correspondingly induce apoptosis and expression of NF-κB.Results MTT method confirmed that combination could inhibit corresponding effect effectively,after 24h and 48h(A 0.489±0.020→0.298±0.096,0.799±0.043→0.311±0.018,P<0.01).Flow cytometry had the strongest detection properties,and cell cycle was blocked in G2/M phase.NF-κB expression was(63.8±5.6)%after cisplatin effect detection with immunohistochemical technique,that was(23.2±2.3)%after combined effect,(P<0.01).Conclusion Quercetin enhances cisplatin-induced apoptosis of colon cancer HT-29 cells,the mechanism may be activating expression of NF-κB through inhibiting,suggesting that combination of quercetin and cisplatin is expected to be applied in tumor chemotherapy.
