二甲双胍通过作用丝状伪足抑制宫颈癌细胞的侵袭和迁移的研究

摘  要:目的探讨二甲双胍对宫颈癌细胞侵袭、迁移能力以及丝状伪足形成的影响。方法实验细胞(Hela)分为对照组和二甲双胍处理组。利用CCK8法检测细胞增殖;流式法检测细胞周期和凋亡;利用Transwell和划痕实验法检测细胞侵袭和迁移能力;用异硫氰酸荧光素-鬼笔环肽染色细胞骨架,观察二甲双胍对HeLa细胞丝状伪足形成的影响。结果10 mmol/L的二甲双胍处理HeLa细胞24 h时,不会抑制细胞增殖,对细胞周期分布和细胞凋亡率均无影响(P>0.05)。Transwell和划痕实验结果证实二甲双胍对HeLa细胞的侵袭、迁移能力均有抑制作用(P<0.01)。二甲双胍可显著降低HeLa细胞丝状伪足的数量和长度。结论10 mmol/L的二甲双胍处理HeLa细胞24 h时可显著抑制其侵袭和迁移能力,机制在于抑制了宫颈癌细胞丝状伪足的形成。Objective The experimental Hela cells were divided into the control group and the MET treatment group.Cell proliferation was detected by CCK8(cell counting kit-8)method;Cell cycle and apoptosis were detected by flow cytometry;Cellular invasion and migration ability were detected by transwell and scratch tests;Cytoskeleton was stained with fluorescein isothiocyanate-phalloidin to observe the effect of MET on the formation of filopodia in HeLa cells.Results The proliferation of HeLa cell was not inhibited after 10 mmol/L MET treated for 24 hours,there was no significant effect on cell cycle distribution and cell apoptotic rate;Transwell chamber and scratch test showed that MET significantly inhibited the invasion and migration of HeLa cells(P<0.01);MET can also significantly inhibit the formation of filopodia in HeLa cells.Conclusions MET 10 mmol/L can significantly inhibit HeLa cell invasion and migration after treated for 24 hours,and the mechanism is in inhibiting the formation of filopodia in cervical cancer cells.
