摘  要:荷叶粗粉用60%乙醇回流提取,经石油醚脱脂,再用聚酰胺柱色谱分离得到不同部位的黄酮提取物。通过比较50%乙醇洗脱部位的黄酮提取物和市售减肥药奥利司他(Orlistat)对胰脂肪酶的抑制作用,得到它们的半抑制率IC50分别为0.0076,0.0048mg/mL。通过Lineweaver-Burk图和Dixon图的特征,表明该抑制剂对胰脂肪酶的抑制呈非竞争性抑制,抑制常数Ki=0.0147mg/mL,米氏常数Km=42.2mg/mL。The powder of lotus leaf was extracted with 60% ethanol completely,and then the mixtures were degreased using petroleum ether.Finally,the various fractions of flavonoids were separated and purified from the extractions by polyamide column chromatography.The effects of 50% ethanol eluted flavonoids extraction and orlistat on the enzyme activity of pancreatic lipase were studied.It was found that the half-inhibition concentrations (IC50) of them were 0.0076 and 0.0048 mg/mL,respectively.Based on the Dixon plo...
