中药补骨脂化学成分及磷酸二酯酶Ⅴ抑制活性测定

摘　　要：经过先后用石油醚、乙酸乙酯及正丁醇对中药补骨脂样品进行萃取,并对所得萃取液用HPLC及1HNMR进一步分离和鉴定,得出其5种组分化合物。通过文献的检索和比较,5种化合物的分子结构也得到证实。这5种化合物依次为补骨脂素、异补骨脂素、补骨脂定、新补骨脂异黄酮及补骨脂异黄酮。还测定了不同有机溶剂萃取液及5种化合物各自的磷酸二酯酶Ⅴ(PDE5)抑制活性。所得试验结果表明,补骨脂中药的主要活性成分是补骨脂素、异补骨脂素及补骨脂定。Five chemical compounds,namely psoralen, isopsoralen,psoralidin,neobavaisoflavone and corylin,were extracted from fructus psoralea with petroleum ether,ethyl acetate and n-butanol successively,the extracts were separated and identified by HPLC and 1HNMR.By checking up with the literatures,molecular structures of the 5 compounds were confirmed.Individual PDE5 inhibition activity of each of the extracts in different organic solvents,at concentration level of 10-6mol·L-1 were determined, and as proved by the r...
