重度碱烧伤视网膜一氧化氮合酶及一氧化氮变化与凋亡的研究

摘  要:目的研究大鼠角膜重度碱烧伤后视网膜诱导型一氧化氮合酶(iNOS)及一氧化氮(NO)的变化与细胞凋亡的关系。方法SD大鼠制作角膜重度碱烧伤模型,在烧伤的不同时间点用酶法检测视网膜组织中iNOS,NO含量的变化,TUNEL法检测视网膜凋亡细胞,并与正常大鼠相对照。结果正常组视网膜中未测到iN-OS含量,仅测到少量NO,重度碱烧伤后大鼠视网膜中iNOS,NO含量均从1d升高,并于7d达最高,30d恢复正常,与正常组比较差异有显著性(P<0.05);重度碱烧伤后3d,7d,15d在视网膜神经节细胞可见不同程度的TUNEL阳性细胞,7d组凋亡阳性表达最强,与其他各组比较差异有显著性(P<0.01)。结论角膜重度碱烧伤后视网膜组织细胞存在凋亡,凋亡可能与iNOS,NO表达有关。Objective To investigate the expression of inducible nitric oxide synthase(iNOS),nitric oxide(NO)in retina after severe corneal alkali burn,and to observe their roles in the apoptosis of retinal cells.Methods The models of severe corneal alkali burn were induced with natrium hydroxydatum.Normal rats were used as control.The NO and iNOS's level was measured by colorimeter and retinal apoptotic cells were stained by TUNEL in chronologicaol order after burn.Results Negative iNOS and microamount NO were measured in the control group.The quantity of iNOS and NO in the retina gradually increased from 1 to 7 days after burn,then decreased which had a significant difference with control groups(P<0.05).The TUNEL(+)cells were found in ganglion cells layer on the 3rd,7th,15th days and attain vertex on the 3rd day,which had significant difference with other groups(P<0.01).Conclusions Some cells in retina tissue will apoptosis after severe alkali burn,which maybe associated with the expression of iNOS and NO.
