摘 要:目的建立大鼠脉络膜新生血管(Choroidal neovascularization,CNV)动物模型,研究临床治疗方法在新生血管发生过程中的作用机制。方法应用氪激光对30只大鼠实验性视网膜进行光凝,功率360mW,光斑直径50(m,曝光时间0.05s,建立CNV动物模型。光凝后3d、7d、14d、21d和28d进行荧光造影,光镜下对苏木伊红染色切片进行CNV形态学评价。采用逆转录聚合酶链反应(Reverse transcription-polymrase chain reaction,RT-PCR)检测视网膜组织VEGF mRNA的变化。结果光凝后7d,FFA光凝斑盘状荧光素渗漏,证实CNV形成,并可持续至28d。光凝后1周可见新生血管。RT-PCR检测证实VEGF mRNA在CNV形成过程中的变化趋势。结论氪激光可诱导BN大鼠产生CNV,持续时间长,病理结构稳定。取视网膜组织进行RT-PCR检测VEGF mRNA,方法可行性强,重复性好。适于作临床治疗方法相关机制的研究。Objective To establish an experimental Choroidal neovascularization(CNV)model in BN rats,which could exhibit high reproducibility and to evaluate the expression of vascular endothelial growth factor(VEGF).Methods 30 rats were received krypton laser.3 days,1 week,2 weeks,3 weeks and 4 weeks after photocoagulation,fundus fluorescein angiography(FFA)and histopathologic examination were performed.Total RNA were abstracted from retinal and choroidal tissue and the mRNA of VEGF was detected by RT-PCR.Results FFA showed flurescein leakage from 1 week to 4 weeks after photocoagulation.Subretinal new vessel(NV)could be observed histopathologically 1 week after photocoagulation.The mRNA of VEGF were significantly increased during 4 weeks by RT-PCR.Conclusions Kryton laser photocoagulation can be successfully used to produced CNV experimental model in the BN rat.CNV pathologic structure possesses stability and lasts long.RT-PCR exhibit high reproducibility and feasibility.This study gave us a effective way to evaluate the mechanism of CNV.
