摘 要:目的探讨亚低温对大鼠局灶性脑梗死后凋亡细胞及谷氨酸转运体(GLT-1)和亲代谢型谷氨酸受体(mGluR2/3)表达的影响。方法88只大鼠随机分为亚低温组、常温组、对照组。制备一侧永久性大脑中动脉阻塞(pMCAO)模型。亚低温组应用贴敷式局部亚低温治疗仪将病灶侧脑部温度降至(33±0.5)℃并持续3h,常温组保持全身温度在(37±0.5)℃。在脑缺血后3h、6h、12h、24h、3d、7d用Western印迹法检测各组病灶周围大脑皮质GLT-1和mGluR2/3表达;在脑缺血后6h、24h、3d、7d用末端标记(TUNEL)法检测凋亡细胞。结果亚低温组在脑缺血后24h、3d,常温组在脑缺血后24h、3d、7d脑梗死灶周围凋亡细胞与脑缺血后6h比较差异有统计学意义(均P<0.01)。在脑缺血后24h、3d、7d,亚低温组凋亡细胞明显少于常温组(均P<0.01)。与对照组相比,亚低温组大鼠脑缺血后3h、6h、12hGLT-1表达增加,24h、3d、7d减少;常温组大鼠脑缺血后3hGLT-1表达增加,24h、3d、7d减少(均P<0.05)。在脑缺血后6h、12h、7d,亚低温组GLT-1表达显著高于常温组(均P<0.05)。与对照组相比,亚低温组和常温组在脑缺血后3h、6h、12h、24h、3dmGluR2/3表达均明显增加(均P<0.05)。在脑缺血后3h、6h、24h、3d,亚低温组mGluR2/3表达显著高于常温组(均P<0.05)。结论亚低温能促进脑缺血后星形胶质细胞mGluR2/3和GLT-1的表达,减少脑梗死后神经元凋亡。Objective To investigate the effects of focal mild hypothermia on apoptotic neurons and the expression of glutamate transporter(GLT-1)and metabotropic glutamate receptor(mGluR2/3)after cerebral ischemia in rats.Methods Eighty-eight adult rats were randomly divided into mild hypothermia group,normothermia group and control group.Unilateral permanent middle cerebral artery occlusion(pMCAO)was induced.In mild hypothermia group,the brain temperature of occlusive side was decreased and maintained for 3 h at(33±0.5)℃ with Sticking Focal Mild Hypothermia Instrument.The body temperature of normothermia group was maintained at(37±0.5)℃.3 h,6 h,12 h,24 h,3 d,7 d after cerebral ischemia,the expression of GLT-1 and mGluR2/3 around infarction cortex were determined by Western Blot.6 h,24 h,3 d,7 d after cerebral ischemia,TUNEL method was performed to label neuronal apoptosis.Results 24 h,3 d after cerebral ischemia of mild hypothermia group,24 h,3 d,7 d after cerebral ischemia of normothermia group,the apoptotic neurons around infarction core markedly increased comparing with 6 h after cerebral ischemia(all P<0.01).24 h,3 d,7 d after cerebral ischemia,the apoptotic neurons of mild hypothermia group were less than that of normothermia group(all P<0.01).Comparing with control group,expreession of GLT-1 in mild hypothermia group increased at 3 h,6 h,12 h,then decreased at 24 h,3 d,7 d after cerebral ischemia;expression of GLT-1 in normothermia group increased at 3 h,then decreased at 24 h,3 d,7 d after cerebral ischemia.6 h,12 h,7 d after cerebral ischemia,expression of GLT-1 in mild hypothermia group were much more than that of normothermia group(all P<0.05).3 h,6 h,12 h,24 h,3 d after cerebral ischemia,expression of mGluR2/3 in the two groups markedly increased.3 h,6 h,24 h,3 d,expression of mGluR2/3 in mild hypothermia group were markedly more than that of normothermia group.Conclusion Mild hypothermia may enhance the astrocytic expression of GLT-1 and mGluR2/3 and reduce neuron apoptosis following cerebral ischemia.
