摘  要:目的探讨血红素氧合酶-1(HO-1)在乳腺良、恶性组织中的表达及意义及其与血管生成的关系。方法分别用RT-PCR、免疫组化及Western blot蛋白印迹杂交检测正常乳腺组织10例;乳腺增生症18例;乳腺纤维腺瘤20例;乳腺癌25例组织中的HO-1mRNA、HO-1蛋白表达,同时免疫组化检测FⅧ,并探讨HO-1在乳腺肿瘤组织中表达的意义及其与血管生成的关系。结果乳腺癌组织中的HO-1mRNA表达(0.375 5±0.062 8)明显高于正常乳腺(0.119 9±0.081 0)、乳腺增生(0.150 21±0.047 9)及乳腺纤维腺瘤组(0.183 4±0.054 1)(t=2.299 2,P<0.01);乳腺良、恶性肿瘤组织中的HO-1蛋白表达(23.461 7±7.013 6,34.271 2±4.342 3)均高于正常乳腺(4.274 2±0.821 57)及乳腺增生组(6.252 5±1.003 2)(P<0.05)。正常乳腺、乳腺增生、乳腺纤维腺瘤到乳腺癌组织的MEA逐渐增多(P<0.01),HO-1蛋白表达与MEA存在线性相关,y=454.66+106.67x,相关系数r=0.8476(t=2.487 3,P<0.02)。结论HO-1mRNA、蛋白表达与乳腺良性病变及癌变存在相关性,HO-1蛋白与血管生成存在相关。Objective To investigate the expression and significance of heme oxyenase-1(HO-1) in benign and malignant breast tissue,and it′s association with angiogenesis.Methods HO-1mRNA and protein in 10 normal,18 hyperplasia,20 adenoma and 25 carcinoma of breast tissue were evaluated by RT-PCR,immunohistochemistry and Western blot respectively,FⅧ-related antigen was examined by immunohistochemistry too,and the relationship between HO-1 and angiogenesis in breast tissue was discussed.Results Expression of HO-1mRNA in breast carcinoma tissue(0.375 5±0.062 8) was higher than that in normal breast tissue((0.119 9)±0.081 0),breast hyperplasia(0.150 21±0.047 9) and breast adenoma(0.183 4±0.054 1)(t=2.299 2,P<(0.01)),HO-1 protein expression in benign and malignant breast tumor tissue(23.461 7±7.013 6,34.271 2±4.342 3) were higher than those in normal(4.274 2±0.821 57) and hyperplasia breast tissue(6.252 5±1.003 2)(P<0.05),and at the same time,the angiogenesis in normal tissue,hyperplasia,adenoma and carcinoma of breast were gradually increased(P<0.01).There was linearity relation between expression of HO-1 and MEA in breast tissue(y=454.66+106.67x)(r=0.8476,t=2.487 3,P<0.02).Conclusion HO-1mRNA and protein expression were associated with benign and malignant breast tissue,and HO-1 protein was associated with angiogenesis..
