宫颈组织临床样本HPV16-E6的阳性率检测

摘　　要：目的探讨HPV-16感染与宫颈癌的关系。方法采用免疫组化SP法分别检测宫颈癌浸润癌23例、慢性炎症25例和正常对照组26例宫颈组织临床样本HPV16E6的阳性率检测及宫颈癌组织中HPV16-E6的表达。结果慢性炎症宫颈上皮细胞杂交阳性率为8.0%(2/25);CIN阳性率为22.7%(5/22);癌变组织阳性率为56.5%(13/23)。其中,I期阳性率为44.4%(4/9),II期阳性率为55.6%(5/9),III期阳性率为80.0%(4/5);正常对照阳性率为3.9%(1/26)。提示HPV16-E6的表达与宫颈癌的发生发展密切相关,随着病变程度的恶化,HPV16-E6的阳性率逐渐增加。将宫颈癌上皮细胞阳性百分率与慢性宫颈炎症比较,具有统计学差异(χ2=19.327,P<0.001),CINs阳性率与慢性宫颈炎症比较也具有统计学差异(χ2=8.0912,P=0.00283),宫颈癌上皮细胞阳性百分率与CINs阳性率相比较,具有统计学差异(P=0.00233)。结论HPV16-E6的表达与宫颈癌的发生发展密切相关,随着病变程度的恶化,HPV16-E6的阳性率逐渐增加。[Objective] To discuss the relationship between HPV-16 infection and cervical cancer. [Methods] immunohistochemical SP method was used to detect the expression of HPV16-E6 and it′s positive rate in 23 cervical infiltrating carcinoma patients, 25 chronic inflammation patients and 26 normal control. [Results] HPV16-E6 positive rate in chronic inflammation cervix endothelial cell is 8% (2/25); in CIN is 22.7% (5/22); in cancerization tissue is 56.5% (13/23) [among it, positive rate in stage 1 tissue is 44.4% (4/9); in stage II tissue is 55.6% (5/9); in stage III tissue is 80% (4/5)]; and in normal control is 3.9% (1/26), It suggest that the expression of HPV16-E6 is correlated with the occurrence and development of cervical cancer. As the process get worse, the positive rate of HPV16-E6 increased gradually. When compare the positive rate in cervical cancer endothelial cell with in chronic cervicitis, the result has statistics variance (χ2=19.327, P =1.2×10-5<0.001); when compare positive rate in CINs with chronic cervicitis also has statistics variance (χ2=8.0912, P =0.00283); the positive rate in cervical cancer endothelial cell with which in CINs has statistics variance (P =0.00233). [Conclusion] The expression of HPV16-E6 is correlated with the occurrence and development of cervical cancer. As the process get worse, the positive rate of HPV16-E6 increased gradually.
