摘 要:目的探讨重组质粒PGL3-DF3-DTA对乳腺癌移植模型的治疗作用。方法裸鼠皮下接种MCF-7人乳腺癌细胞,建立人乳腺癌移植模型。用质粒PGL3-DF3-DTA,质粒PGL3-DF3和生理盐水分别给裸鼠移植瘤瘤内多点注射。观察肿瘤大小和组织形态学变化,并用流式细胞仪检测肿瘤细胞凋亡改变。结果重组质粒PGL3-DF3-DTA组裸鼠乳腺癌生长受到明显抑制,抑制率为50.08%,肿瘤细胞的凋亡率增加。结论重组质粒PGL3-DF3-DTA对裸鼠人乳腺癌移植瘤的生长具有明显抑制作用,为乳腺癌基因治疗提供了新的方法。[Objective] To study the effect of recombinant plasmid PGL3-DF3-DTA on breast cancer xenografted on a nude mouse model. [Methods] Balb/c nude mouse was injected subcutaneously with 107 human breast cancer cell line MCF-7. Normal sodium, plasmid PGL3-DF3 and recombinant plasmid PGL3-DF3-DTA were respectively injected into the breast cancer xenograft on the nude mouse with some kind of dose every three days. The tumor volume was measured before every injection. The mice were killed after five injections. The histopathological changes of the tumor were observed by HE stain. Tumor cell were analyzed with flow cytometery to detect apoptosis and cell cycle. [Results] Growth of human breast cancer was significantly inhibited in the recombinant plasmid PGL3-DF3-DTA therapy group as compared with the normal sodium group (P <0.01) and the growth inhibitory rate was 50.08%. Apoptosis rate was higher than that of the normal sodium group and the plasmid PGL3-DF3 group. [Conclusion] Our finding suggests that recombinant plasmid PGL3-DF3-DTA results in marked inhibition of human breast cancer growth in nude mice. It may be a novel gene therapy approach for human breast cancer.
