内毒素致伤大鼠肝组织SSeCKS mRNA水平的研究

摘　　要：目的观察内毒素/脂多糖(LPS)致伤大鼠肺组织中Src抑制的蛋白激酶C的底物(SSeCKs)mRNA的表达变化。方法SD大鼠腹腔注射LPS建立肝损伤模型,依据注射时相和剂量不同随机分组。用Real-timePCR法检测肝组织SSeCKSmRNA水平的表达情况。结果不同剂量LPS注入腹腔后8小时均可引起肝脏SSeCKS的表达变化,且呈现一定的剂量依赖关系。5mg/kgLPS腹腔注射后肝脏SSeCKSmRNA水平的表达量达到最高,此后虽然增加LPS剂量但也不能诱导SSeCKSmRNA水平表达量的增加;5mg/kgLPS腹腔注射后肝脏SSeCKS的表达变化呈现时间依赖关系,LPS注射后1小时肝脏SSeCKSmRNA水平表达增高,12小时达到最高水平,此后SSeCKS一直维持高水平。结论LPS引起肝脏SSeCKSmRNA表达呈时间和剂量依赖性变化,提示SSeCKS与LPS所致肝损伤相关。Objective To investigate the changes of the expression of SSeCKS in the injured liver tissue of rat induced by LPS.Methods The LPS-induced liver tissue injury model of rats was set up intraperitoneally.The injured rats were grouped randomly according to different dosages of LPs and time phases.The expression of the mRNA of SSeCKS in the liver tissue of different group was determined by Real-time PCR.Results SSeCKS expression change was followed dosage and time dependent manner.The expression of SSeCKS in mRNA level arrived the peak in 5mg/kg LPS group;the expression level of SSeCKS increased after i.p.LPS,after 12 hour it arrived the peak,even at 24hour its still stayed high.Conclusions The results provides an evidence that SSeCKS expression change depends on different dose and time of LPS challenged liver.This indicates SSeCKS is related with LPS injured liver.
