摘  要:利用产D-氨基酸氧化酶(DAAO)和戊二酰基-7-氨基头孢烷酸酰化酶(GLA)的高酶活重组大肠杆菌,对双酶催化制备7-ACA的工艺进行了研究。提出了一条游离细胞反应与膜分离耦合的7-ACA生产新工艺:用游离细胞进行催化,用超滤膜进行细胞和产物的分离。结果发现用中空纤维超滤膜可以将反应体系中的细胞和催化产物分离,分离得到的细胞可重复使用;分离得到的反应液经一步结晶,7-ACA纯度为93.1%,结晶收率为51%。The recombinative strains of D-amino acid oxidase(DAAO) and glutaryl-7-amino-cephalosporanic acid acylase(GLA) were used to produce 7-aminocephalosporanic acid(7-ACA) by hydrolysis of cephalosporin C,and the catalyzing conditions of these two enzymes were optimized.A novel process of free cell catalyzing combined with hollow fiber membrane separation was developed for production of 7-ACA.The free cells were used in catalysis while the hollow fiber membrane was applied to entirely separate the cell and the product.The product can totally pass through the membrane,and the cells were reusable.The purity of 7-ACA prepared by this process could reach 93.1% with a yield of 51%.
