摘 要:应用酸性聚丙烯酰胺凝胶电泳(A-PAGE)、十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)和简单重复序列(simple sequence repeal,SSR)标记,对穗数型多分蘖小麦品种川农16与大穗型寡分蘖小麦品系H461生化及分子水平上的遗传差异进行了检测。结果表明,川农16与H461间至少有10条醇溶蛋白差异带,其高分子量谷蛋白亚基组成分别为(1,20,5+10)和(2*,7+8,2+12)。在小麦21条染色体上的72个SSR 位点上共检测N95个等位变异,每一个位点所检测到的等位变异数目为1至2个,平均1.3个;其中,23对SSR 引物(31.9%)能够揭示川农16与H461间的差异。Genetic variations of seed storage protein composition and SSR between a wheat cultivar Chuannong 16 with more spikes per plant and a uniculm wheat line H461 were evaluated. The composition of gliadins, analyzed by the acid polyacrylamide gel eletrophoresis (A-PAGE), indicated that 10 bands were polymorphic between Chuannong 16 and H461. High molecular weight glutenin subunit composition of the two genotypes were identified by sodium dodeeyl sulphate-polyacrylamid gel electrophoric (SDS-PAGE), indicating that Chuannong 16 has 1,20,54-10 subunits and H461 has 2 * ,7 + 8,2 + 12 subunits. A total of 95 alleles were detected on 72 SSR loci. On each SSR locus, 1 to 2 alleles were detected, with the mean of 1. 3 alleles. Twenty-three out of 72 SSR markers (31. 9%) were polymorphic between the two genotypes.
