摘 要:为了确定类球红细菌转硒培养的最佳条件 ,研究了无机硒的加入浓度、时间以及分批补料培养对菌体生长和转硒效率的影响。实验表明 ,无机硒的浓度低于 1× 10 -5mol/L时 ,对类球红细菌的生长基本没有影响 ,并能将6 3.9%的无机硒转化为有机硒。转硒的最佳时间是在接种后 12h左右 ,此时转硒效率最高。实验还表明 ,分批补料培养可以提高菌体浓度 ,可使转硒效率和绝对量增加。体内试验表明 ,用 5mL/kgbw和 10mL/kgbw剂量的含硒类球红细菌灌养小鼠 ,可以使其全血GSH Px酶活性提高 2 0 .9%和 2 5 .5 % ,使其血清丙二醛 (MDA)含量降低2 1.0 %和 2 3.2 %。In order to find the best conditions for the growth of Selenium-containing Rhodobacter sphaeroides No.1, we have studied the effect of the concentration, the time of adding Se() and the fed-batch on the growth of R.sphaeroides No.1 and the efficiency of transferring Se. As the concentration of Se() is less 1×105mol /L ,the growth of No.1 is almost not been suppressed, it can transfer 63.9% of Se() to organic Se.The best time of adding Se is the 12h after the inoculation and the efficiency is the highest. We also found that the fed-batch can enhance the concentration of thalli and increase the efficiency and the absolute value of transferring Se. Through the experiment in vivo, We have found that feeding mice with the 5ml/kgbw and 10ml/kgbw of Se-containing thalli of R.sphaeroides can enhance the enzyme activity of GSH-Px reached to 20.9% and 25.5%, and decrease the value of MDA 21.0% and 23.2%. ;
