紫外线诱发人晶体上皮细胞凋亡及其抑制的研究

摘  要:目的 :研究不同剂量的紫外线对体外培养的人晶体上皮细胞凋亡率的影响 ,观察维生素E及人参皂甙 (Rg1)对其干预作用。方法 :同一紫外线光源下一固定位置 ,测得该点的功率为 3 0 6μw/cm2 ,按照不同的照射时间分别为6、12、18min ,对第 2~ 3代人晶体上皮细胞进行照射 ,用流式细胞仪检测其凋亡率 ,透射电镜观察凋亡细胞的超微结构的改变。结果 :不同的照射时间 ,凋亡率分别为 :2 0 %、5 3 % ,81% ,透射电镜下见核物质离散 ,胞膜隆凸及特征性的凋亡小体。经维生素E处理的人晶体上皮细胞 (HLEC)凋亡率为 3 5 % ,(对照为 5 3 % ,P <0 .0 1) ,经人参皂甙处理的HLEC凋亡率为 3 9% (P <0 .0 1)。结论 :紫外线照射诱发晶体上皮细胞凋亡 ,凋亡率随着紫外线剂量的增加 (照射时间的延长 )而增加。维生素E、人参皂甙可部分抑制紫外线诱发的晶体上皮细胞凋亡。Objective: To investigate the apoptosic rate of human lens epithelial cells induced by ultraviolet(UV)radiation of different dose and the inhibitory effect of Vit E and Rg1. Methods:Conflurent monolayers of cultured human lens epithelial cells were exposed to ultraviolet lamps emiting predominantly UVA radiation for 6,12, and 18 minutes. The apoptosic rates of cells were surveyed separately with flow cytometry, and the microscopultrastructure alteration of apoptosic cells was observed with ectron microscopy. Apoptosic rates of the cells cultured with or without Vit E of 10 μg/ml and Rg1 of 50 μg/ml were surveyed. Results: At 6',12',18', the apoptosic rate was 20%, 53%, and 81%, respectively. The apoptosic rate of cells in culture medium without Vit E or Rg1 increased as compared with that of cells cultured with Vit E(P<0.01) or Rg1(P<0.01). The cells were damaged by UV radiation, which were characterized as separated nucleic materials, bulged membrane, and appeared apoptosis body. Conclusion: UV can induce the apoptosis of the human lens epithelial cells. The apoptosic rate increases with increased dose of UV radiation. VitE and Rg1 could inhibit the apoptosis induced by UV radation.
