摘　　要：目的探讨SU11248对结肠癌细胞系SW480中趋化因子受体CXCR4表达的影响。方法采用RT—PCR技术和免疫细胞化学染色法检测结肠癌细胞系SW480细胞中CXCR4 mRNA和蛋白的表达。采用MTT法检测SU11248对SW480细胞增殖的影响。用Westernblot检测SU11248作用后CXCR4蛋白及相关的信号调控分子的表达。结果SW480细胞中CXCR4 mRNA和蛋白呈阳性表达;SU11248对SW480细胞增殖和CXCR4表达有明显抑制作用;SU11248对调控CXCR4表达的相关的信号通路有抑制作用。结论SU11248可抑制结肠癌细胞增殖,降低CXCR4在结肠癌的表达。Objective To investigate the effect of SU11248 in the expression of chemokine receptor CXCR4 in colorectal carcinoma cell lines.Methods To detect the expression of CXCR4 receptor in the cell line with immunohistochemistry and RT-PCR.To detect the influence of cell proliferation treated by SU11248 with MTT assay and to detect the expression of CXR4 protein and the correlative signaling molecule after SU11248 treated with Western-Blot method.Results CXCR4 mRNA and correlative protein have Positive expression....
