摘　　要：目的检测卵巢癌细胞株SKOV3中骨桥蛋白(OPN)的表达,探讨OPN对卵巢癌细胞侵袭潜能的影响。方法pcDNA3.1(-)/OPN重组质粒转染SKOV3细胞,以空质粒转染作对照。采用RT-PCR及Western blot法检测OPNmRNA和蛋白表达水平,并用基质凝胶侵袭实验检测两组细胞侵袭力。结果重组质粒转染SKOV3细胞后OPNmRNA和蛋白表达明显升高。侵袭实验证实重组质粒转染组细胞侵袭力明显增加,差异有统计学意义(P<0.05)。结论OPN对卵巢癌细胞的侵袭潜能有促进作用。Objective To investigate the expression of Osteopontin(OPN) and explore its effect on the invasiveness of human ovarian carcinoma cell line SKOV3.Methods Human ovarian carcinoma SKOV3 cells were transfected with plasmid pcDNA 3.1(-)/OPN and cells transfected with a mock plasmid served as controls.RT-PCR and Western-blot were used to determine the expressions of OPN mRNA and proteins.Cell invasiveness was measured by the matrigel invasion assay.Results Expressions of OPN mRNA and protein were significantly i...
