摘　　要：目的研究羟基喜树碱两种构型在肿瘤组织中的分布和比例关系。方法采用HPLC-FLD法,内标法测定。结果肿瘤组织中羟基喜树碱与内标喜树碱峰分离良好,内源性杂质不干扰样品峰的测定,酯型羟基喜树碱的最低检测浓度为2ng·mL-1,浓度范围在2.6～2600ng·mL-1间呈线性相关,相关系数r=0.9993,样品的加样回收率分别为96.04%～98.58%,日内和日间变异均小于10%。结论本方法具有专属性,灵敏,可靠,能够满足肿瘤组织中羟基喜树碱药动学及相关研究的需要。Objective To study the distribution and ratio of the two forms of hydroxycamptothecin in mice tumor tissues.Methods HPLC-FLD method was used with internal standard method.Results Good isolation of hydroxycamptothecin from the internal standard(camptothecin) peak was obtained in tumor tissues, endogenous impurities did not affect the peak of the sample.The lowest detection concentration of lactone hydroxycamptothecin was 2ng·mL-1.A linearity was shown in the concentration range of 2.6～2600ng/mL.The regressio...
