发色多肽用于前激肽释放酶活性测定的研究

摘　　要：目的:弄清N-t-BOC-Leu-Ser-Thr-Arg-pNA作为激肽释放酶测定的发色底物的基础参数并建立血浆(前)激肽释放酶测定的程序。方法:以激肽释放酶及其他相关因子的制品及缺乏血浆测定其Km,max,Ki,回收率,对比试验,并确定最适测定条件。结果:该发色底物受激肽释放酶作用的Km和Vmax分别为235μmol/L和337 nmol(pNA).S-1.U-1(KK);该底物的酰基水解活性不依赖因子、、和蛋白C,而明显依赖于激肽释放酶,且对AT-和LBTI的抑制均不敏感。用该多肽建立了血浆激肽释放酶活性测定法,该方法与传统方法无论是活性测定或抗原定量均有很强的可比性,线性范围6.25%~300.0%。结论:B3644作为激肽释放酶测定的发色底物,具有良好的敏感性和选择性,是建立PK活性测定的理想发色底物。Objective:To identify the specifications of N-t-BOC-Leu-Ser-Thr-Arg-pNA as a chromogenic substrate in the determination of kallikrein,and then establish the determination procedure.Method:By determining the peptide s Km,Ⅴmax,Ki,the recovery rates,and,comparison tests,for kallikrein and other related plasma factors,to decide the optimized measuring condition.Results:The kallikrein Km and Ⅴmax for this peptide were determined to be 235 μmol/L and 337 nmol.s-1.U-1,respectively.Its amidolysis was independent of...
