摘  要:目的 :观察帕金森病 ( PD)大鼠模型黑质区诱导型一氧化氮合酶 ( i NOS)的基因表达 ,探讨其与神经损伤的关系。 方法 :用 RT-PCR法、免疫印迹分析及还原型辅酶 ( NADPH)黄递酶组织化学法观察 PD大鼠黑质区i NOS的基因表达、神经胶原纤维酸性蛋白 ( GFAP)和 NADPH黄递酶阳性细胞的变化 结果 :PD大鼠受损侧黑质有明显的 i NOS基因表达 ,未损侧仅有轻度表达 ;对照组大鼠两侧黑质均无 i NOS基因表达。PD大鼠受损侧黑质GFAP表达较未损侧明显增加 ,受损侧黑质较未损侧出现增多的 NADPH黄递酶阳性胶质样细胞。 结论 :i NOS基因在黑质反应性星形胶质细胞上的表达可能参与Objectives:To observe the expression of inducible nitric oxide synthase(iNOS) gene and explore its role in dopaminergic neuron damage in rat model of Parkinson disease(PD). Methods:The expression of iNOS gene and glial fibrillary acid protein(GFAP) in the substantia nigra in rat model of PD were detected by RTPCR and Western blots analysis respectively. Results:High levels of iNOS mRNA were detected in the substanstia nigra of lesioned side in rat model of PD, while only low levels of iNOS mRNA in unlesioned side; no expression of iNOS were detected in both side of the substanstia nigra in control rat. The GFAP protein expression of lesioned substantia nigra in rat model of PD displayed a remarkable increase compared to unlesioned side, and an increased NADPH-diaphorase positive glial cell appeared in the lesioned substantia nigra. Conclusions:The expression of iNOS gene in reactive astrocytes may participate in the processes of dopaminergic neuron damage in the substanstia nigra of rat model of PD.
