摘 要:目的探讨乳腺癌(breast cancer)患者外周血和乳腺组织中CD4+CD25+、Foxp3 mRNA和蛋白水平表达情况,初步阐明二者在乳腺癌患者免疫调节中的作用和意义。方法选取乳腺癌患者25例,15例正常乳腺组织作为对照。采用流式细胞术检测乳腺癌患者外周血中CD4+CD25+Treg的表达水平。RT-PCR和Western blot分别用于检测患者癌变组织中Foxp3 mRNA和蛋白表达水平。结果相对于正常对照组,各组乳腺癌患者外周血中CD4+CD25+Treg表达水平明显升高(P<0.05);同时,乳腺癌各组中Foxp3 mRNA和蛋白表达水平均明显升高(P<0.05,P<0.01)。结论CD4+CD25+Treg和其标记分子Foxp3可能参与了乳腺癌中机体的免疫调节过程,其二者表达水平的检测对于乳腺癌临床判断和进一步的治疗具有一定指导意义。Objective To investigate the expressions of CD4+CD25+ regulatory T cell(Treg) and Foxp3 in peripheral blood and mammary gland tissues of breast cancer patient with different pathological types and illuminate the effect and the significance in immunological regulation. Methods Total of 35 breast cancer tissue samples including 25 infitrating ductal carcinoma,4 infiltrating lobular carcinoma,4 colloid carcinoma,and 2 carcinoma medullare were selected according to 6th Edition of AJCC Cancer Staging Manual for breast cancer. Meanwhile,15 normal mammary gland tissue samples were selected as control. Flow cytometry was employed to determine the expression level of CD4+CD25+ Treg in peripheral blood of the patients. RT-PCR and Western blot were used to determine Foxp3 mRNA and protein expression level,respectively. Results CD4+CD25+ Treg expression levels in breast cancer tissue was increased obviously compared with normal control (P<0.05 ). Foxp3 mRNA and protein levels in various groups were increased significantly (P<0.05,P<0.01). Conclusion CD4+CD25+ Treg and its labeled molecule Foxp3 might involve in the genesis,development,and immunological regulation of breast cancer. Determination of their expression levels might benefit the clinical judgement and further therapy for breast cancer.
