摘 要:目的探讨脑源性神经营养因子(brain-drived neurotrophic factor,BDNF)对抑郁症大鼠海马神经元前体细胞微管相关蛋白Doublecortin(DCX)表达的影响,为BDNF如何影响神经元前体细胞的增殖、迁移提供实验依据。方法成功建立抑郁症大鼠模型后,向脑室注射0.5μgBDNF,取脑组织行冰冻切片。采用免疫组织化学和免疫荧光技术观察DCX阳性细胞表达情况及表达部位。结果相对于未注射BDNF的抑郁症大鼠,注射组大鼠海马区域DCX阳性细胞数明显增多,且与未注射组存在明显差异(P<0.05)。结论BDNF可能通过调控抑郁症大鼠海马神经元前体细胞微管相关蛋白DCX的表达,从而影响抑郁症时神经元前体细胞的分化和迁移。Objective To investigate the effects of brain-drived neurotrophic factor (BDNF) on the expression of neuronal precursors microtubule-associated protein doublecortin in hippocampus of depressed rats and provide experimental evidence for the effects of BDNF on the proliferation and migration of neuronal precursors. Methods Depressed rat model was established,and 0.5 ug of purified BDNF was injected into rat cerebral ventricle for preparing frozen section. Immunohistochemistry and immunofluorescence were employed to observe the positive expression level and expression site of Doublecortin (DCX). Results Positive cell number of DCX in hippocampus region of BDNF injection group was increased compared with that of non-injection group (P<0.05). Conclusion BDNF might affect the differentiation and migration of neuronal precursors via regulating the expression of DCX in hippocampus of depressed rats.
