摘 要:目的探讨基质细胞衍生因子1(SDF-1)及其受体CXCR4在大肠癌组织中的表达及其临床意义。方法应用免疫组织化学SP法检测60例大肠癌、23例大肠管状腺瘤及22例正常肠黏膜组织中SDF-1和CXCR4的蛋白表达情况。结果大肠癌组织中SDF-1和CXCR4阳性表达情况与大肠管状腺瘤及正常肠黏膜组织比较,差异均有统计学意义(P<0.05);且大肠管状腺瘤组织中SDF-1和CXCR4阳性表达情况与正常肠黏膜组织比较,差异亦有统计学意义(P<0.05)。不同浸润深度及有无淋巴结转移的患者,大肠癌组织中SDF-1及CXCR4阳性表达情况间差异均有统计学意义(P<0.05)。大肠癌组织中SDF-1的表达与CXCR4的表达呈正相关(r=0.436,P<0.01)。结论大肠癌组织表达SDF-1及其受体CXCR4,且二者在大肠癌发生、发展、浸润、转移中发挥着重要作用。Objective To evaluate the role and clinical significance of SDF-1 and its receptor CXCR4 in development,infiltration and metastasis of colorectal carcinoma.Methods The levels of SDF-1 and CXCR4 protein expression were detected by SP immunohistochemical method in 60 patients with colorectal carcinoma(CC group),23 with colorectal adenoma(CA group) and 22 with normal colorectal mucosa(NCM group).Results CC group was significantly different from groups CA and NCM in positive expressions of SDF-1 and CXCR4(P<0.05),which was the highest in CC group among the three(P<0.05).Stronger expressions of SDF-1 and CXCR4 proteins were observed in CD group,as compared with NCM group(P<0.05).The positive expressions of SDF-1 and CXCR4 were closely correlated with the depth of tumor infiltration and metastasis of lymph node(P<0.05),but not with genders and ages of patients and differentiation of cancer.The positive expression of SDF-1 was positively correlated with that of CXCR4 in colorectal carcinoma(r=0.436,P<0.01).Conclusion SDF-1 and CXCR4 are important factors in development,infiltration and metastasis of colorectal carcinoma.
