胃癌发生发展中β-连环蛋白异常表达与Wnt传导通路激活的关系

摘　　要：目的探讨β-连环蛋白(β-catenin)异常表达与Wnt传导通路激活在胃癌发生发展中的作用及关系。方法选取62例胃癌患者的癌组织,并选取35例胃部距癌组织2～5cm处的癌旁组织和22例切缘正常的胃黏膜组织作为对照,应用免疫组织化学方法检测Wnt传导通路的信号蛋白(Wnt2)和β-catenin的表达情况;在上述标本中选取30例胃癌组织、15例癌旁组织和15例正常胃黏膜,应用流式细胞技术检测Wnt2和β-catenin的表达情况。结果β-catenin在正常黏膜、癌旁组织及癌组织胞膜中的表达强度呈下降趋势,而在胞质中的表达强度呈增强趋势,3组间差异有统计学意义(H胞膜=45.60,P=0.000;H胞质=23.83,P=0.000);Wnt2在正常黏膜、癌旁组织及癌组织中的表达强度呈上升趋势,3组间差异有统计学意义(H=23.83,P=0.000)。β-catenin在正常黏膜、癌旁组织及癌组织中的荧光指数(FI)分别为(1.00±0.06)、(0.97±0.05)、(0.88±0.07),3组的表达量间差异有统计学意义(H=26.66,P=0.000)。Wnt2在正常黏膜、癌旁组织和癌组织中的FI分别为(1.00±0.11)、(1.08±0.65)、(4.95±1.54),3组的表达量间差异有统计学意义(H=44.30,P=0.000)。结论在胃黏膜癌变过程中Wnt信号通路处于激活状态,可能影响了β-catenin在细胞内的分布,二者共同影响胃癌的发生、发展进程。Objective To investigate the correlation between the abnormal expressions of β-catenin and Wnt signaling pathway activation in gastric carcinogenesis.Methods Cancer tissues were taken from 62 gastric carcinoma cases,and adjacent tissues (2～5 cm from cancer tissues) from 35 and normal incisal margin gastric mucosa tissues from 22 were taken as controls.The expressions of Wnt2 and β-catenin were detected by immunohistochemistry;30 cases of cancer tissues,15 adjacent and 15 normal gastric were taken from the above-mentioned specimens to determine Wnt2 andβ-catenin expressions.Results The expression of β-catenin exhibited a down trend in gastric cancer tissues,adjacent and normal gastric,and a up trend in cytoplasm,the difference between 3 was significant(Hmembrane=45.60,P=0.000;Hcytoplasm =23.83,P=0.000);Wnt2 expression increased gradually in normal gastric tissuses,adjacent and cancer,the difference between 3 was significant(H=23.83,P=0.000).β-catenin FI values in normal gastric tissues,adjacent,cancer were 1.00±0.06,0.97±0.05,0.88±0.07,respectively,the difference was significant(H=26.66,P=0.000),and Wnt2 FI values were 1.00±0.11,1.08±0.65,4.95±1.54,respectively,the difference was significant(H=44.30,P=0.000).Conclusion In the process of gastric carcinogenesis Wnt signaling pathway is in activation status,probably influencingβ-catenin distribution in cells.Both effect the process of gastric carcinogenesis.
