高效液相色谱法测定余甘子中多种氨基酸的含量

摘　　要：建立了高效液相色谱法测定余甘子中多种氨基酸含量的方法,采用 ZORBAX EclipseAAA 4.6×150mm,5μm色谱柱;OPA 柱前衍生氨基酸;荧光检测器检测;多步梯度洗脱;可在25min 内分离出16种氨基酸。本方法快速,样品用量少,灵敏度高,线性范围宽,适用于其他食品中氨基酸含量的测定。A method for determination of amino acids in phyllanthus emblica L.by high performance liquid chromatography(HPLC)with fluores- cence was developed.HPLC was performed on a ZORBAX Eclipse AAA column(4.6×150mm,5μm).more than 16 amino acids were separated within 25min by using pre-colurnn derivatization with O-phthalsldehyde and a multi-step gradient with two solvents.The method is fast、sensi- tive、accurate and has a linear range from 20-400pmol/μL.
